Study On Preparation And Quality Standard Of Three Dispensing Granules Of Chimonanthus Nitens,Catsia Tora And Nelumbinis Folium

Chimonanthus nitens is geoherbs of JiangXi,is the plant of Calycanthaceae Chimonanthus.Medicinal part is the dry leaf,which has the effects of anti-cold,relieve a cough,expectorant,anti-bacterial,anti-inflammatory,anti-pyretic,analgesic,expel toxin,losing weight,lowering blood pressure,heatstroke prevention,stopping diarrhea,aidding digestion and so on.Semen Cassiae is the mature dry seeds of leguminous plants of Cassia obtusifolia L.or Cassia tora L.,it is sweet,bitter,salty,slightly cold,has the effects of clearing heat,eyesight,laxative,protecting liver,reducing blood lipid,lowering blood pressure,enhance immunity,antibacterial,diuresis,diarrhea,anti-oxidant,anti-platelet aggregation and so on.Nelumbinis Folium is the dried leaves of the Nymphaeaceae Nelumbo nucifera Gaertn.,it is bitter,flat,owned by the liver,spleen,stomach,clear heat and wet,cooling blood to stop bleeding,used for summer wet,summer heat,thirsty,irritation,diarrhea,blood heat,metrorrhagia.Pharmacological research shows that Nelumbinis Folium has the effects of losing weight,anti-bacterial,anti-oxidation,anti-aging,anti-virus,anti-inflammory and anti-allergic.Traditional Chinese herbal medicine is the raw material of traditional Chinese medicine dispensing granules,dispensing granules is made by modern Chinese medicine technology,including extraction,concentration,separation,drying,granulating,packaging and testing.It can be used alone,also can be used for clinical Chinese medicine prescription dispensing,has the advantages of strong resistance,less dosage,composition complete,fast and convenient,high efficiency,safety,sanitation,etc.,so the study of Chimonanthus nitens dispensing granules,Semen Cassiae dispensing granules,Nelumbinis Folium dispensing granules has important significance,the research carried out the following work:Single factor experiment and orthogonal experiment were used to optimize the extraction process of Chimonanthus nitens,Semen Cassiae and Nelumbinis Folium respectively.Single factor experiment was used to optimize the molding process of the above 3 kinds of Chinese herbal medicines.Single factor experiment and L₉?3⁴?orthogonal design were carried out in the extraction process,with the index component transfer rate and herbal extract rate as indexes,to inspect 3 factors of extraction times,heating time,amount of water,so as to determine the optimum water extraction processes of 3 kinds of Chinese herbal medicines.After verification,the corresponding water extraction process was scientific and reasonable.Single factor experiment was carried out in the molding process to inspect 4 factors of supplementary materials,the quantity of supplementary materials,ethanol concentration and ethanol dosage,so as to determine the optimum molding processes of 3 kinds of Chinese medicine dispensing granules.After verification,the corresponding molding process was reasonable and feasible.Finally,the preparation processes were determined by the above work.The quality standards of Chimonanthus nitens dispensing granules,Semen Cassiae dispensing granules,Nelumbinis Folium dispensing granules were established based on the systematic research of identification,examination and determination of the content.Qualitative identification was carried out by TLC.Rutin,scopoletin,scoparone and isofraxidin in Chimonanthus nitens dispensing granules were identified;Aurantio-obtusin and chrysophanol in Semen Cassiae dispensing granules were identified;Nuciferine,rutin and quercetin in Nelumbinis Folium dispensing granules were identified;The corresponding spots were clearly visible,and the negative samples had no interference.The items of examination included character,particle size,moisture,melting,and the results showed that all indexes were eligible.By optimizing the extraction conditions and chromatographic conditions of 3 kinds of Chinese medicine dispensing granules,to establish the methods for determination of the active compounds in the 3 kinds of Chinese medicine dispensing granules by HPLC,the corresponding methods are simple,accurate and reliable,which provide reliable basis for the establishment of quality standards and quality control of 3 kinds of Chinese medicine dispensing granules.In this topic,Chimonanthus nitens decoction and dispensing granules fingerprints,Semen Cassiae decoction and dispensing granules fingerprints,Nelumbinis Folium decoction and dispensing granules fingerprints were compared respectively.The results showed that the corresponding decoction and dispensing granules in species composition almost unchanged,so the study of 3 kinds of dispensing granules is scientific and reasonable,has a certain significance of research.The preliminary stability tests were performed for three batches of 3 kinds of dispensing granules respectively.After three months of accelerated stability tests,the results of three batches of 3 kinds of dispensing granules indicated that 3 kinds of dispensing granules were stable.Instrument analysis is an important method of chemical analysis,and has broad application prospects.In the studies,modern instrument analysis technology was used to study the preparation and quality standard of Chimonanthus nitens dispensing granules,Semen Cassiae dispensing granules,Nelumbinis Folium dispensing granules,such as UV,HPLC and so on.These studies provide reliable basis for the development,utilization and the quality evaluation of 3 kinds of Chinese medicine dispensing granules.
