Cinnamomum camphora var.borneol is an important tree species of aromatic oil,which are rich in multiple volatile oil,especially a high content of natural borneol.Because of the trend of short supply in market,it is crucial to look for Cinnamomum camphora var.borneol clone with high content of natural borneol and excellent cold resistance as a female parent to produce a lot of high quality seedlings.Moreover,it has a great help for the reasonable development and utilization of Cinnamomum camphora var.borneol resources.This article mainly studied on superior individual selection and container cutting propagation.By measuring the content of natural borneol in leaves,composition of volatile oil content and resistance physiological indicators,we get the superior individual.Container cutting experiment is made through different cutting substrates,hormone treatment,lignified degree and cutting length,so as to select the most suitable solution for cuttings rooting.The results are as follows:1.Preliminary screening of superior individual: The differences of natural borneol from100 plants of borneol camphor leaves is obvious by using steam distillation.33 plants were selected,according to the content of natural borneol in leaves were over 1.0 percent,respectively,Z67?Z55?Z82?Z10?Z29?Z32?Z92?Z60?Z19?Z84?Z56?Z86?Z93?Z96?Z48?Z77?Z100?Z41?Z13?Z2?Z45?Z8?Z5?Z17?Z20?Z9?Z23?Z68?Z30?Z95?Z64?Z74?Z4.Among them,Z67 is the highest,which reached 1.66 percent.2.Intermediate screening of superior individual: collect and treat essential oil in leaves that extracted by steam distillation.30 compounds are identified by detecting essential oils in leaves of 33 plants that selected from primary screening by GC-MS.Of them,only nine important volatile oil contents which had higher economic value were analyzed.Then,screen out 15 plants that total volatile oil over 1.50% and 9 important volatile oil over 1.00%,respectively,Z2,Z5,Z9,Z10,Z17,Z19,Z30,Z32,Z48,Z55,Z60,Z67,Z77,Z82,Z86.3.Ultimate screening of superior individual: cold resistance of 15 plants that selected from intermediate screening was studied.Firstly,measure the hardiness indexes,such as relative conductivity,MDA content,soluble sugar content,soluble protein content,Pro content,SOD activity and CAT activity.Then,analyze them with fuzzy membership function,also combined with winter morphology observation.Finally,select 6 plants as the final superior individuals which have strongest cold resistance,respectively,Z17,Z19,Z32,Z48,Z67 and Z86.4.The influence of different matrix on container cuttings rooting of Cinnamomum camphora var.borneol is different.Matrix A8,namely peat soil,perlite,river sand and vermiculite mixed by 1:1:1:1,is better than other matrix formula.The average rooting rate is82.33%,average root length is 10.59 mm,and average root number is 7.48.So it is the most suitable matrix formula for container cuttings rooting of Cinnamomum camphora var.borneol.5.Hormones and concentration have different effects on container cuttings rooting of Cinnamomum camphora var.borneol.The most suitable concentration of each hormones for container cuttings rooting is 400 mg·L-1IBA?400 mg·L-1NAA?200 mg·L-1ABT-1#.However,there are significant differences in different hormones for container cuttings rooting.Of them,200 mg·L-1 ABT-1# is the best.The average rooting rate is 89.33%,average rooting length is10.52 mm,and average rooting number is 7.47.6.Significant differences exist in different lignifications for container cuttings rooting of Cinnamomum camphora var.borneol.The average rooting rate of half-lignified branches is88.67%,average rooting length is 10.35 mm,and average rooting number is 7.48.7.Cutting length also plays different roles in container cuttings rooting of Cinnamomum camphora var.borneol.Among them,the average rooting rate of 7 cm branches is the highest with 89.33%,and average rooting number is also the most with 7.37;9 cm branches is the longest average rooting length by 10.35 mm.The average rooting rate of 5cm branches is88.67%,which is second only to 7 cm branches.Above all,superior individuals of Cinnamomum camphora var.borneol clone will be screened out by high content of natural borneol and excellent cold resistance;meanwhile through testing,we get the most appropriate scheme of container cuttings rooting.In order to meet the demand for market,the superior individual can be used as female parent to obtain lots of asexual reproduction and high quality off springs.So it can provide a basis for making scientific and reasonable use of the Cinnamomum camphora var.borneol.
