Study On Cinnamomum Camphora Presl New Germplasm With Cold Resistance Through Genetic Engineering

Cinnamomum camphora Presl is a kind of broad-leaved evergreen arbor and has big crown and a few fallen leaves. Cinnamomum camphora Presl is a kind of high Practical application ornamental tree,and could be as street tree,shade tree in courtyard.But it could not adapt the cold weather,New variety can be get from genetic engineering method. So the Cinnamomum camphora Presl can lives in west-China.The embryo callus from Cinnamomum camphora Presl were transformed with AmEBP1 and AmGS mediated by Agrobacterium.,according to sequence analysis and cold tolerance text.The results:⒈To screen medium suitable for Cinnamomum camphora Presl's growth and differentiation.Lateral buds were strongly effected by NAA. 0.1mg/LNAA had good effects on lateral buds. Best induction rate can be get when the BA:2,4-D is 1:4.best cytokininin was BA, best phytohormone was NAA in the same medium. The suitable mediumfor propagation was MS+5.0mg/LBA+1.0mg/LIBA.⒉The embryo callus from Cinnamomum camphora Presl were transformed with AmEBP1 and AmGS mediated by Agrobacterium.The results of PCR assay and sequeneing indicated that target gene has been transferred into Cinnamomum camphora Presl.⒊The results of the cold tolerance text showed that the death rate of the transgenic vitro plants was lower than CK 67.0% under -17℃,2h。In the range of 0～-16℃,relative conductivity was rise,and tmembrane relative penetration was rise too, LT50 was 1.12℃lower than CK. Cold resistance of Cinnamomum camphora Presl turn strong.
