This experiment collected two suspected pseudorbies virus(PRV) infection cases from two pet hospitals in Nanchang, Jiangxi province. After autopsy and pathological analysis, polymerase chain reaction(PCR) detection was carried out for the two cases. Meanwhile, one of cases was selected for isolation and identification test in vivo and in vitro, and the two strains' virulence genes, gD and gE, were studied for understanding the molecular characteristics and evolutionary relationship. Additionally, serological investigation of canine pseudorbies(PR) was carried out in Nanchang area in this experiment. The results were as follows.1 The PRV gE fragment, which size was 346 bp, was successfully amplified from the two dogs' cerebrums. And the two strains were named PRV JX-1-2014 and JX-2-2015, respectively. The virion was observed as solid or hollow in infected PK-15 cells by electron microscope. The strain's TCID50 attained 10-4/0.1 mL when titrated in PK-15 cells culture. Typical symptom of PR could be observed in adult rabbits after inoculating the JX-1-2014 strain.2 The gD and gE genes of PRV, which size were 1208 bp and 1770 bp, were successfully amplified. Compared with domestic and international PRV strains, the two Jiangxi strains have got variations, and were relatively distant from the international PRV strains, while had close relationship to the domestic strains. Additionally, more closely relationship was found between the strains and domestic strains after 2011.3 The results of serological investigation demonstrated that antibody positive of PRV and PRVgE were found in the dogs surveyed. The total positive rate of PRV antibody was 36.8%(74/201).Conclusions: The diseased dogs were actually infected by PRV, and the isolated strains had some degree of variations. At present, canine PRV is considered to be pandemic risk in Nanchang area.
