Gastrointestinal granules for fish feed consist of these Chinese herb extracts:Amur Corktree Bark, Cortex Fraxini and wood anemone. The gastrointestinal granules was employed as our research object to study the best dosage and application technique of Chinese medicine_to prevent disease in the aquaculture.Ointment recovery rate and the content of Berberine hydrochloride was taken as the index for investigation in the orthogonal experiment. The results showed that the optimized preparation conditions for the gastrointestinal granules was as follow:add 10 times water to these three traditional chinese medicine, decoction three times, each time lasting an hour. The anointed rate and the content of Berberine Hydrochloride is the highest.The reverse phase high performance liquid chromatography (RP-HPLC)and ultraviolet spectrophotometry were used to determine the content of Berberine hydrochloride which is the gastrointestinal granules active principles. Compared with ultraviolet spectrophotometry method, the precision, reproducibility, stability, and recovery rate of RP-HPLC method was better. The RP-HPLC method with simple sample pretreatment, high sensitivity and accuracy, may serve as a way of quality stantard draving up.The stability of the gastrointestinal granules were investigated by accelerated experiment and room temperature experiment and light influencing-factor test. The results showed that:after 10 days light influencing-factor test,6 months acceleration test and 24 months of storage at room temperature, each index of the sample such as character, content changed less. So its stability was satisfactory.Acute toxic experiments are carried out to test the safe concentration of mix the gastrointestinal granules feeding for japanese eel, soft-shelled turtle ricefield eel and Penaeus vannamei Boone. The test result showed that the safe concentration of mix feeding was more than 3g/kg. According to the result, both the preventive dose:1-2 g per kg of feed, and the therapeutic dose:2?3 g per kg of feed, using 5?7 days continuously are safe.In drug sensitivity trial, the gastrointestinal granules has certain inhibition on most common pathogenic bacteria for infective gastroenteritis. The minimal inhibitory concentration (MIC) of the compound Chinese medicinal granules against Flexibacter columnaris, Vibrio anguillarum, Aeromonas hydrophila were all 0.625?g/mL, And the MIC of the Aeromonas sobria was 1.25?g/mL.In pharmacodynamics trial, to the acute or chronic enteritis induced by bacterial infection, adding 2-3 g per kg this granules in grass carp feed, using 5-7 days continuously, the mortality of the control group was over 70%, but the mortality rate was decreased to less than 50% in the experiment group, and the protection rate of the medicine was 30% or so on average.It was proved in production trial that the gastrointestinal granules was effective in against the acute or chronic enteritis induced by bacterial infection, and the enteritis, enterelcosis, enterorrhagia caused by toxic substances secreted by harmful algal, and the gastrointestinal discomfort, feeding down, dyspepsia caused by phytotoxicity factors.
