| Taihu goose is the high quality germplasm resource in China,with high productivity,strong disease resistance,good feeding,high rate of returning,early sexual maturity,weak broodiness,less illness,fresh meat and other characteristics,but the scale of farming is declining sharply because of its slow growth,light weight,the exotic breeds greater impact and inbreeding leading to the degeneration of variety.In order to make full use of the germplasm resources and understand the early growth,slaughter performance and other economic traits,the research is looking for the SNPs and its main economic traits related to or haplotype,provide technical support for the Taihu goose molecular marker assisted breeding,so as to provide theoretical basis for the protection and exploitation of the species.We researched on principal component analysis of Taihu goose body weight and body size traits,analysised the polymorphism of two Taihu goose candidate genes,Adiponectin and Ghrelin gene,related to the production performance by direct sequencing method and use the least squares method to analyze its correlation with growth traits and carcass traits.The results showed that:(1)The 9 index of body size traits and body weight on the Taihu goose were mea-sured.Between male and female geese comparative study shows:in higher male goose body ruler index and its relationship with body weight as compared to the goose;male goose semi submerged length and chest width are extremely significant positive correlation(P<0.01)and the goose semi submersible length and hip width are extremely significant positive correlation(P<0.01).Later on in the production practice by semi submersible long to understand male goose breast development and goose pelvis development,have very good guidance.Male goose can be extracted from the five principal components F1,F2,F3,F4,F5,and their cumulative contribution rate for 86.780%,each principal component the maximum load in order to keel length,body slanting length,shank length,chest width,hip width and therefore should be at least select one of the above five indicators of male goose growth traits for breeding.Goose can also be extracted from the five principal components F1,F2,F3,F4,F5,the cumulative contribution rate 87.586%,from its eigenvectors can be seen,each principal component the maximum load in turn for hip width,tibia length,chest width,chest depth,so at least we should select one of the above four indicators of goose growth traits in breeding.(2)SNPs on most of the area of Adiponectin and the part region of Ghrelin of Taihu goose by direct sequencing technique were respectively scanned,and it was found:8 SNP locus of Adiponectin gene:C155T,G244A,C252T,C422T,A428G,A429G,C431T,G605A9 SNP locus of Ghrelin gene:C76T,C145G,C266T,A289G,A373G in intron 2 and T9C,A72G,A127C,A311G in intron 3.The test result shows that all the seat of P values were less than 0.05,indicating that each locus data from the same Mendelian populations in Hardy Weinberg equilibrium.(3)Linkage disequilibrium analysis showed that between C155T,G244A,C252T,C422T and G605A and between A428G,A429G and C431T of Adiponectin gene are in perfect linkage disequilibrium state and between C76T and A289G,C145G and T9C,C266T,A373G and A72G of Ghrelin gene were in strong linkage disequilibrium;A127 also reached the significant linkage disequilibrium with C266T,A373G,A72G respectively and.(4)The genetic correlation analysis is showed that:Adiponectin gene C155T,A428G site of Taihu goose leg muscle rate are significantly different(P<0.01).C155T site on tibia length and leg muscle weight had significant effect(P<0.05).A428G site on diving length,chest width and leg muscle weight had significant effect(P<0.05);Ghrelin gene C76T sites of plagioclase is significantly different(P<0.01),to 6 weeks of age,body weight and keel has significant effect(P<0.05);sites C145G and T9C for 1 week old,weight,chest deep,tibia length had significant effect(P<0.05);T266C,G373A,A72G,A127C sites have significant effect on the width of chest(P<0.05);site G289A of plagioclase is significantly different(P<0.01).The keel has significant effect(P<0.05);site A311G on body slanting length,chest width has significant effect(P<0.05). |
