Sophora japonica as an important native species in China has a long history of cultivation,it has been a good street trees and landscape trees shrubs.In recent years,its value is increasingly prominent,whether ecological value,ornamental value,economic value,or the value of the humanities have aroused strong concern,therefore it is more and more in-depth study.In this paper,we studied the Sophora japonica seedlings of 4 years.Observe and measure the height beyond the branches.The seedling populations of Sophora japonica genetic variation analysis and individual plant selection was established,tissue culture.The study yielded the following results:1.Genetic diversity and selecting elite individual plant(superior tree).The size of the diameter at breast height of the tree high discrete degree,varies greatly from the other.This article will research status at origin of the health Samuel lam traits on the basis of genetic variation,by using the method of small standard,we selected 10 excellent single strains of good traits,The rate of admission was 3.3 percent,the good single tree was 111% higher than the average of the forest,and the diameter of the chest was over the average of 147% of the forest,and over 220% of the average.They all grow strong,have no branches,and the trees are graceful.2.Sophora japonica stems start culture medium was the best,MS+100ml/L Coconut water+0.5mg /L 6-BA+1.0mg/L NAA,The inducement rate was 75%;Optimum medium for subculture was MS + 100ml/L Coconut water + 1.5mg/L 6-BA + 0.15mg/L NAA,Differentiation rate of embryogenic callus reached 86%;Using the 1/2MS + 120ml/L Coconut water + 0.2mg/L NAA + 0.5g/L AC as the root regeneration medium,the rate of rooting can reach 11%.Transplant the test tube in 3-4 months,the matrix's best choice is humus soil and a beaker can reduce the loss of moisture,Increase the survival rate of transplant.
