Hydropericardium syndrome is caused by Fowl Adenovirus group I type 4,its second name is Angara disease.The disease was a global outbreak in many country,such as Iraq,India,Chile,southern and central United States,Canada,Hungary,Poland,Korea,Russia,Japan.In recent years,Liaoning,Shanxi,Shandong,Hebei,Inner Mongolia,Hunan,Hubei,Jilin,Jiangsu and other places also reported the disease.The disease is causing huge economic losses to the aquaculture industry.Two different strains were used in this study.One from a broiler breeder farm in Tianjin Province,65-week-old broiler breeder.Another one from a layer chicken farm in Fushun,Liangning Province,11-days-old layer.DNA was extracted from the liver of the suspected lesion and amplified by specific primer PCR.Liver lesions was grinding postprocessing,repeated freezing and thawing three times after centrifugation the supernatant fluid was sterile filtrate.The viral fluid were inoculated into the allantoic cavity of 7 day old SPF chicken embryos,discard 24h nonspecific death of embryo,collected within 24h-168h of dead embryo allantoic fluid and non dead embryo,continue to inculated in the 6 day old SPF chicken embryo on blind transfer the 3 generation.The third generation of allantoic fluid was collected by aseptic treatment,and the filtrate was detected by asepsis.The filtrate was inoculated into the control group by the mouth of 1 day old SPF chickens and was killed at the age of 7 days.The test of nine blood physiological indexes.,including blood total protein(TP),albumin(ALB),alkaline phosphatase(AKP),alanine aminotransferase(ALT),urea nitrogen(BUN),uric acid(UA),creatinine(CRE),lactate dehydrogenase(LDH),creatine kinase(CK).Results:from two group of disease in chicken liver were amplified successfully in Ankara virus genome segment.Two strains of isolates were blind to the third generation of chick,chick embryo can lead to growth retardation,curled up,bleeding in the T group,Tianjin strain inoculated chicken embryos after 6D after the death of.SPF chicken F were inoculated with virus(Fushun strain then,starting from 4d)the phenomenon of death.Histological observation showed that the isolated two strains of virus can cause a variety of myocardial cells,liver tissue cells and renal tubular epithelial cells and the change of particles,vesicular degeneration and necrosis.The damage indicated that detection of blood biochemical indicator,two strains of virus can be isolated from chicken in Ankara the decreased content of total protein change and albumin in serum of chickens,the slight increase in lactate dehydrogenase content of urea nitrogen,creatinine and creatine kinase content increased significantly,while alkaline phosphatase,ammonia lyase and uric acid with alanine aminotransferase,the quantity is greatly increased.It proves that both strain T and stain F can seriously affect liver fanction,renal function and heart function.Conclusion:the two strains of chicken Angara virus,named T strain and F strain,two strains of the virus may cause the embryo and chick characteristic pathological injury and changes in heart,liver and kidney function.
