| Schisandra,Aralia elata,Acanthopanax as the food and medicine homemade woody wild vegetables,as the embryo development is not complete,there are serious seed dormancy phenomenon.And the use of cutting propagation is not easy to take root,resulting in such trees buds on the wild seedling breeding difficulties.In this experiment,different stratification temperature,different exogenous substances deal with the seeds of three wild vegetables,using different exogenous hormones,different substrates,different temperature cutting cuttings,to study the effects of different methods on seed germination and cutting effect.Aiming at providing reference for the rapid propagation of tree buds and wild vegetables.Through research,we get the following conclusions:(1)The temperature stratification treatment is beneficial to the germination of three kinds of buds,and the higher the temperature stratification time,the better the effect of the variable temperature stratification treatment.Schisandra chinensis,Aralia elata and Acanthopanax senticosum were used in the high temperature stratification of 12 ℃ and then combined with 4 ℃ low temperature stratification(Tw3、Tw4、Tw5;Tl3、Tl4、Tl5;Tc3、Tc4、Tc5)were effective in improving the seed embryo occupancy rate and seed germination rate;In the case of consistent number of layers,the embryo occupancy rate and seed germination rate were significantly increased with the higher temperature stratification time(shortening the later period low temperature stratification time).Among them,Schisandra chinensis was treated with supercooling at 12 ℃ for 60 days and then at 4 ℃ for 30 days.Aralia elata and Acanthopanax Garcusta were all treated with 12 ℃ high temperature 90 d combined with late 4 ℃ low temperature 60 d temperature stratification treatment effect is the best.(2)A certain concentration of exogenous substances-Gibberellin(GA3),Cytokinin(6-BA),and copper sulfate effectively increased the seed germination rate of three kinds of buds.The germination rate of 200 mg / LGA3 was the highest in the other treatments,and the germination rates were 61.67%,66.22% and 63.48%,respectively,which were significantly higher than those of Schisandra chinensis,Aralia elata and Acanthopanax senticosus.(3)A mixture of exogenous hormones-indole butyric acid(IBA),naphthalene acetic acid(NAA),indole butyric acid and naphthalene acetic acid could significantly improve the survival rate and root activity of three kinds of tree buds,500 mg / lIBA + 500 mg / l NAA mixture(treatment B11)the best.The cuttings,cutting survival rate and root activity of three kinds of buds were studied byusing different concentrations of IBA,NAA,IBA: NAA = 1: 1(concentration setting of 250,500,1000,2000 mg / l)the increase of three kinds of exogenous hormones showed the trend of increasing first and then decreasing.In the process of cutting root regeneration,the root activity increased with the cutting time.(B11)was the highest survival rate and root vigor,and the cutting effect was the best.The cutting efficiency was the highest.The survival rates of cuttings of Schisandra,Aralia elata and Acanthopanax senticosus were 51.61%,42.09% and 48.33%,respectively.(4)Three kinds of tree buds Wild vegetables cutting appropriate substrate for the peat:vermiculite = 1: 1,the appropriate ground temperature of 25 ℃. |
