Licorice is Perennial herbaceous plants of Leguminosae and Glycyrrhiza Linn.,which is known as the king of Chinese medicine and has important medicinal value.It is one of the most commonly used traditional Chinese medicines in China.However,at present,Because of liquorices resource scarcity,there are still many problems to be solved,such as improving the quality,breeding and cultivation of licorice and so on.DNA methylation is one of the most significant epigenetic regulatory mechanisms in eukaryotic cell genome,which regulates the function of the genome without altering the DNA sequence,allowing the gene to be expressed as needed,and keeping silence when it is not needed.In plants,DNA methylation is mainly involved in plant gene expression regulation,regulating the growth and development of plants.In order to maintain the normal growth and development,plants can regulate gene expression through methylation and demethylation in different environmental conditions and different stages of development.In this paper,the methylation-sensitive polymorphism(MSAP)was used to analyze the DNA methylation of licorice seeds under high voltage electrostatic field stress(18kV/25min).MSAP results showed that the methylation level of CCGG site in licorice dry seeds was about 12.89%in high voltage electrostatic field treatment,which indicates a 0.7%increase when exposed to high voltage electrostatic field,and mainly in full methylation pattern.During the germination process,the DNA methylation level of seeds increased first and then decreased.In the cotyledon phase,the total methylation level was 11.42%in control and 17.48%in high voltage electrostatic field treatment,indicating a 6.06%increase after high voltage electrostatic field stress,mainly in hemi-methylation pattern.During the germination of licorice seeds,methylation and demethylation happened at the same time and showed complexity.In summary,the change of DNA methylation is a complicated process during seed germination.Glycyrrhizin may regulate the expression of the gene by methylation and demethylation,and ultimately determine the growth and development of plants.
