Lepista sordida is a rarely edible and medicinal fungi.The number of the wild fungus is low and it has a high development value,which contributes to its high price.To provide reference for later research on lepista sordida,JZ01 lepista sordida collected from Zhuhai College of Jilin University lab building lawn,By Chemistry and Department of pharmacy Lanying Wang teacher appraisal,named JZ01 lepista sordida.In this paper,the technique of RAPD molecular marker and 16S-23 S rRNA internally transcribed spacer(ITS)were used to gene identification of JZ01 lepista sordida,and research on its antibacterial activity of fermented liquid.With CTAB method adopted,the genomic DNA from five different origins of mycelium and sporocarp of lepista sordida was extracted and used in RAPD amplification.The eight of clear band primpers selected by twelve from Shanghai Sangon Biological Engineering Company were conducted data analysis and PCR amplification with five samples,obtaining forty six articles of amplified fragments.Using NTSY software,the genetic similarity(GS)of the sample was caculated to obtain the similarity matrix about the materials.The range of the sample's genetic similarity(GS)is: 0.2826-0.9130,and the rangeability is 0.6304.The bigger the genetic similarity(GS)is;the closer the genetic relationship is,and vice versa.According to this law:The genetic similarity(GS)between sample 4 and sample 5 is the biggest,0.9130,which shows the genetic relationship between the two is more close and it has less genetic difference.However,the genetic similarity(GS)between sample 3(JZ01)and sample 4,5 is the smallest,0.2826,which shows the JZ01 lepista sordida along with sample 4 and 5 has a farther genetic relationship and greater difference,which means they share a more higher genetic diversity.Compared with the five sequences of ITS,the results show that.The genetic distance is: 0.00460-0.06752,and the rangeability is 0.06292.The genetic distance between sample 4 and sample 5 is the smallest,0.00460,which shows the genetic relationship between the two is more close and it has less genetic difference.However,the genetic distance between sample 1 and sample 5 is the biggest,which shows the genetic relationship between the two has a farther genetic relationship and greater difference,which means they share a more higher genetic diversity.The cluster analysis results demonstrated that the genetic relationship between five kinds of samples are identical with the result of RAPD,shows using these two methods to identify the genetic relationship of lepista sordida is feasible.In the study of fermented liquid of lepista sordida antimicrobial activity,using different organic solvent,the results show the extracelluar small moleules ethyl acetate phase extracted from lepista sordoda has certain inhibition effect on the growth of the selected strains and the effect is relatively good.Especially to serratia marcescens inhibitory is best.However,the antibacterial effect of the small molecules n-butyl alcohol phase is bad,not suitable for use.The extra cellular small molecule of the JZ01 lepista sordida has certain inhibition effect on G+ and G-,which manifests JZ01 lepista sordida has a wide range of antibacterial activity,can be used as a natural antibacterial agent to develop applications.
