In this paper,the chemical constituents of liquid fermentation broth of Lepista sordida and the fruiting-body of Russula sp.were purified by chromatography.Their chemistry structures were identified based on NMR,MS,and other physicochemical data.L.sordida was fermented in the shaking flasks with potato glucose media for52.5 L.The fermentation broth was filtered to remove hyphae.Then the fermenta tion broth was concentrated and extracted with ethyl acetate in equal proportion an d obtained the crude extract(5.6 g).10 compounds(JY11,JY12,JY15,JY17,JY19,JY21,JY24,JY26,JY27 and JY28)were isolated from the fermentation broth of L.sordida by using various column chromatographic techniques over reverse p hase silica gel,Sephadex LH-20 and silica gel.According to the physicochemical data of NMR,MS and UV absorption spectra,their structures of JY11,JY12,JY21,JY24,JY26,JY15,JY17,JY19,JY27 and JY28 were elucidated as 4-hydroxy-3-methyl-1-(2,3,4-trihydroxy-5-(3-hydroxyprop-1-en-2-yl)-6-methylphenyl)butan-2-one,7,8-dihydroxy-5,5,9-trimethyl-3-methylene-3,3a,5,6-tetrahydro-1H,4H-benzo [de]isochro men-4-one,4,9a,10-trimethyldecahydro-5H-5,8-methanobenzo [7]annulene-1,4,5-triol,(3aS,6S,8aR)-3,8a-dimethyloctahydroazulene-2,3a,6(1H)-triol,4,7-dihydroxy-2-(hydroxy methyl)-2,8-dimethyl-3,5-dihydro-s-indacen-1(2H)-one,geosmin,(4aS,5S,8aS)-5,8a-dim ethyloctahydronaphthalene-1,4a(2H)-diol,Lepistamide A,(4-aminophenyl)methanol a nd Lepistatin A.Compounds JY11,JY12,JY21,JY24 and JY26 were new sesquit erpenes through the literature retrieval.200 g dry powder of Russula sp.was extracted with methanol by ultrasonic extraction and concentrated to afford the crude extract 9.6 g.Then the crude extract was partitioned with the equal volume of n-butanol phase and water.n-Butanol phase was concentrated to afford 1.4 g.Then the pH value of water phase was adjusted to 9.0 and extracted with ethyl acetate and n-Butanol.The organic phase was concentrated to afford the organic crude 0.34 g.7 compounds JY02,JY03,JY06,JY07,JY09 and JY10 were isolated from the fruiting bodies of Russula sp by column chromatography over reverse phase silica gel,Sephadex LH-20 and silica gel.According to the physicochemical data of NMR,MS and UV absorption spectra,their structures of JY02,JY03,JY06,JY07,JY09 and JY10 were elucidated as adenosine,9-octadecenoic acid,cytosine,cerebroside,cytidine and mannitol.The experimental results in this paper may shed light on the chemistry foundation of their pharmaceutical values of two edible and medicinal fungi L.sordida and Russula sp.and would underlie the further development.
