Rumex gmelini Turcz.belongs to the family of Polygonaceae.Its root has been used for antifungal,antitumor,antitussive,antiasthma,antivirus and antioxidation.Through the regulation of different plant hormones,Rumex gmelini loose callu was obsained in the 2,4-D 0.5 mg/L,6-BA 3.0mg/L,sucrose 30g/L,culture temperature(25±2)?,light 14h/d,light intensity 1500?2000Lx culture conditions yellow green loose granular callus were selected for Rumex gmelini suspension cells culture.The result indicuted that the initial concentration of inoculation amount was 1g and 2,4-D,0.5 mg/L,6-BA,2.5mg/L,sucrose 30g/,culture temperature of(25±2)?,rotational speed 120rpm,the culture time of 18d were optimal culture condition.The fresh weight cells reached the maximum amount of growth when cultured for 18d,Fresh weight growth rate is 1.0103g.Methods of the endophyte traits,endophyte inoculum,inoculation time,rotate speed and the initial cell concentration,were studied to find out the best method for establishment of the co-culture of Rumex gmelini suspension cells and endophytic fungi.Fungal spore was added to 3-day-cultured plant suspension cell,and continue shaking culture in the dark at(25±2)?,120r/min.
