Candida tropicalis R1,Saccharomyces cerevisiae N2 and Nguyen candida C3 are chosen to produce probiotic,and the bioactivity of three kind of yeast are measured with the method of solid fermentation based on Single factor optimization and Orthogonal optimization(L9(34))in order to confirm the best fermentation time,the best inoculation amount,the best ratio of material to water and the solid Medium of yeast.After all,the most suitable fermentation condition and the optimum culture medium of the 3 kinds of yeast are confirmed,to acquire the product technology of the high production active yeast probiotic.The products are used on animal production application at the same time to verify its feeding effects.The results are as follows:1.In temperature of 30? and condition of natural pH,the best fermentation time of Candida tropicalis RI is 48 hours,the best inoculation amount is 11%,the best ratio of material to water is 1:0.75,the best fermentation time of Saccharomyces cerevisiae N2 is 72 hours,the best inoculation amount is 7%,the best ratio of material to water is 1:1.25,the best fermentation time of Nguyen candida C3 is 48 hours,the best inoculation amount is 11%,the best ratio of material to water is 1:1.25.2.The best carbon source type of solid state fermentation medium on yeasts R1?N2?C3 is glycerol,the best nitrogen source type is carbamide;the optimum solid-state fermentation culture medium formula of yeasts R1,in 8.5 to 1.5 proportion between bran and glycerol(g),carbamide 0.5%,in yeasts N2 the optimum formula is in 9 to 1 proportion between bran and glycerol(g),carbamide 0.5%,potassium dihydrogen phosphate 0.2%,in yeasts C3 the optimum formula is in 8.5 to 1.5 proportion between bran and glycerol(g),carbamide 0.5%,potassium dihydrogen phosphate 0.4%.During experiment of solid state fermentation in optimized fermentation conditions and culture medium formula,the viable count of R1 is 32.94×108cfu/g,and 2.26 times higher than before,the viable count of N2 is 8.6×108cfu/g and 6.32 times higher than before,the viable count of C3 is 2.66×108cfu/g and 1.09 times higher than before.3.The production performance of mutton sheep can be improved by adding complex yeasts probiotic to daily diet,the average daily gain weights of mutton sheep are 164.89g/d and 236.45g/d in experiment during two periods.Compared with control group,the average daily gain weight of mutton sheep increases 48.19%,and gain ratio decreases 39.01%.
