As a functional oligosaccharide,raffinose is a kind of safe and effective excellent bifidobacterium growth enhancing factor,can improve intestines health,enhances immunity and has other pharmaceutical physiological function.Gossypol has pharmacological functions,such as antifertility active and anti-tumor.The main source of raffinose and gossypol is cottonseed meal.The content in cottonseeds has a direct impact on the production efficiency.So,improving the raffinose or gossypol content in cottonseeds is of great significance to the development and utilization of raffinose or gossypol.This study adopted the method of EMS chemical mutagenesis to create mutants in Gossypium arboreum,used High performance liquid chromatography(HPLC)to analysis the change rule of raffinose and gossypol content in G.arboreum mutagenesis progeny,and screened out high-gossypol,high-raffinose and low-gossypol cotton mutants for the use of exploiting natural cotton seeds as biological medicine.The major research contents and results include:1.In order to create high raffinose cotton mutants population,we optimizes technical parameters of EMS mutagenesis in G.arboretum.The results showed that the treatment with 0.6% EMS for 8 h was appropriate for G.arboretum Shixiya 1 mutagenesis.Under the above treatment conditions,field surveys showed that the mutation frequency of agricultural traits of G.arboretum Shixiya 1 was 48.37% and the rate of heritable mutants was 3.00%.The mutants were screened and classified after large-scale mutagenesis of G.arboretum being done,and 5920 M2 generation were harvested.This work had laid a solid material foundation for high raffinose cotton mutants population creatment.2.46 M1 generation Asian cotton mutants and their offspring M2 generation mutants were randomly selected.The raffinose and gossypol were extracted from cottonseeds using physical methods.The contents of raffinose and gossypol were measured by high performance liquid chromatography(HPLC)technique,to analyze their change rule.The statistical results indicated that the contents of raffinose and gossypol in the seeds of Gossypium hirsutum Zhongmiansuo 24 were measured higher than G.Arboretum,the analysis of variances of the contents of raffinose and gossypol in the seeds between different varieties of G.Arboretum were not obvious,and the contents of raffinose and gossypol in the seeds were measured higher than the control materials(Shixiya 1)after EMS treatment.There were 44 high raffinose M2 cotton mutants,and the highest content of raffinose in M2 materials was 55.4567 mg/g,significantly increased 49.10% comparing with the control group Shixiya1.There were 18 high gossypol M2 cotton mutants,and the highest content of gossypol in M2 materials was 10.8041 mg/g,significantly increased 33.54% comparing with the control materals.There were 8 low gossypol M2 cotton mutants,and the lowest content of gossypol in M2 materials was 6.5310 mg/g,significantly reduced 19.27% comparing with Shixiya 1.3.There were 5 high-raffinose and low-gossypol cotton mutants being screened out from 46 G.arboretum M2 generation mutants,according to the reference value of the raffinose content of Gossypium hirsutum Zhongmiansuo 24 and the gossypol content of Gossypium arboreum Shixiya 1.We believe there will be lots of high-raffinose and low-gossypol mutants being screened out from the rest 5870 M2 lines,which will provide rich inheritable material for the study of development and utilization of raffinose.
