Pistacia chiensis Bunge belong to genus Pistacia,the family Anacardiaceae.The deciduous tree has beautiful and lush leaves,is frequently used as an ornamental tree.P.chinensis is also avaluable for timber,medicine and oil production.In recent years,the attention on studying P.chinensis has focused on the economic,landscape and social values of P.chinensis.In this study,the germinated seeds of P.chinensis were used as experimental materials.The defoliated stem-segments of one,year old seedlings with a single bud were used to establish the aseptic culture.The conducted experiments to evaluate salt,drought and cold tolerance of P.chinensis,and the physiological and biochemical indices under the different environments of stress were measured to correlate these activities with stress tolerance in P,chinensis.The main experiment results are following.For in vitro establishment,the best culture medium formula was:1/2 Woody Plant medium(WPM)supplemented with 1.5 mg/L of 6-Benzylaminopurine(6-BA),0.1 mg/L Naphthalene Acetic Acid(NAA),and 0.1 mg/L Indole Butyric Acid(IBA),with rate of 91.7%.For shoot proliferation the best medium was Murashing Skoog medium(MS)supplemented with 3.5 mg/L 6-BA,and 0.2 mg/L NAA.For in vitro rooting,the best medium was 1/2 WPM supplemented with 0.02 mg/L NAA,and 1.0 mg/L IBA.For the stress tolerance ability of P.chinensis were measured under the stress of salt,drought,and cold treatments changes of superoxide dismutase(SOD),peroxidase(POD),catalase(CAT)activities and malonaldehyde(MDA)contents.Compared with the untreated samples,the SOD,POD,and CAT activities changes of the salt treatment samples first went up,then got down.Research showed that P.chinensis were tolerant to salt.Under drought and cold stresses,the activity of SOD,POD,CAT and content of MDA in the leaves of P.chinensis had limited changes,and lower then the untreated samples.P.chinensis had intense drought and hypothermia tolerance.
