Litter size is the most important economic traits in sheep,but the litter size trait heritability is low threshold traits,the traditional methods improvement progresshas been slow.At present,with the emergence of new biotechnology,especially molecular marker technology is widely used in the initial screening of sheep,greatly improving the efficiency and accuracy of selection.This experiment using PCR-SSCP and direct sequencing of BMPR-IB gene and ADAMTS-1 gene genetic polymorphism analysis of the effect of fecundity of sheep,And analysis of litter size in sheep were associated with biological information analysis,The expression of ADAMTS-1 gene in different tissues of sheep was analyzed by real-time fluorescent quantitative technique,and the effect of ADAMTS-1 gene on the transcription level was verified.The research aimed to provide some reference for improving the sheep fecundity.The results are as follows:1 Analysis of Polymorphism at 864 Locus of BMPR-IB Gene CDS Region and Its Relationship with Litter Size in Sheep BreedsGansu Alpine Merino ewes singleton,Gansu Alpine Merino ewes twins,Mongolia ewes singleton,Mongolia ewes twins,Hu ewes multiple births and Small Tailed Han ewes multiple births in six sheep populations,found that the BMPR-IB gene coding region 864 th site T>C synonymous mutation.In Small Tail Han sheep and Hu sheep ewes lamb ewes,Mongolia ewes singleton and Mongolia ewes twins ewes of four populations was only detected in AA;AB genotypes.However,there were three genotypes of AA,AB and BB in Gansu Alpine Merino ewes singleton and Gansu Alpine Merino ewes twins.Hu ewes multiple births and Small Tailed Han ewes multiple births advantage were AA genotype,However,the frequency of AB genotype Gansu Alpine Merino ewes singleton,Gansu Alpine Merino ewes twins,Mongolia ewes singleton and Mongolia ewes twins was slightly higher than that of type AA.The dominant alleles of all sheep populations were A.The distribution of genotypes in different sheep populations was examined,The distribution of different genotypes in 4 sheep breeds was different,Gansu AlpineMerino(singleton,twins)respectively in Small Tail Han sheep and Hu sheep had extremely significant difference(P<0.01),Mongolia sheep(singleton,twins)respectively in Small Tail Han sheep and Hu sheep had extremely significant difference(P<0.01),But the difference in high fecundity sheep breeds sheep and Small Tail Han sheep in between is not obvious(P>0.05).The mutation of the BMPR-IB gene coding region 864 th was different in the different fecundity ewes.Comparison with genotype by use of varieties and littersize after interaction,the difference between varieties and varieties within the larger,distribution difference is not obvious.The study showed that the BMPR-IB gene encoding region 864 mutation is correlated with litter size.2 Analysis of Polymorphism at Partial 3?UTR BMPR-IB Gene and Its Relationship with Litter Size in Sheep BreedsCombined with DNA sequencing using PCR-SSCP technology in Gansu Alpine Merino ewes singleton,Gansu Alpine Merino ewes twins,Mongolia ewes singleton,Mongolia ewes twins and Small Tailed Han ewes multiple births of 5 sheep populations in the 3 'part of the non encoding region were found in non encoding region of 511 st loci and 544 th loci in C>T C>G mutation.Three kinds of genotypes of CC,CT and CG were found in the population of 5 sheep.In Gansu Alpine Merino populations,CT genotype has a significant effect on litter size(P<0.05),while no significant effect of different genotypes in Mongolia ewes singleton,Mongolia ewes twins groups for litter size(P>0.05).3 Differential mRNA Expression of Sheep ADAMTS-1 Gene in Different Tissues and Analysis of Exon 5 PolymorphoismAccording to the Small Tailed Han ewes multiple births,Mongolia ewes singleton,Mongolia ewes twins ADAMTS-1 gene mRNA in kidney,heart,liver,lung,spleen,pituitary,hypothalamus,ovarian tissue expression analysis results show that in the 3 group,the expression of ovary was significantly higher than other organizations(P<0.05),the expression of Small Tailed Han ewes multiple births ovarian is 1.54 times the Mongolia twins sheep ewes,Small Tailed Han ewes multiple is 2.61 timesthe Mongolia ewes singleton.The results show that the ADAMTS-1 gene of mRNA in different groups of sheep ewes lambing trait in ovarian tissue showed large differences in ADAMTS-1 at the transcriptional level for the fecundity of sheep had significant effects.The analysis of the polymorphism of exon 5 of ADAMTS-1 gene,Small Tailed Han ewes multiple births,Mongolia ewes singleton,Mongolia ewes twins in the CDS region 1506 loci C>T,1509 loci G>A,belong to synonymous mutation,three kinds of type named AA,AB,AC,significant analysis three genotypes were not significant on litter size(P>0.05).
