The formation of hair follicles is the result of the interaction between the dermis and the epidermis,and the mesenchymal-epithelial interaction determines the fate of the hair follicle.The hair shaft is produced by the coordination of the dermal papilla mesenchymal cells with the surrounding epithelial stromal cells.In the cyclical development of hair follicles,many signaling pathways are involved,Wnt10 b and FGF18 gene play a key role in the process of hair follicles.In this study,the lateral skin of Tianzhu white yak was used as material,and Histological techniques,Real-time PCR and immunohistochemical techniques were used to control the morphology and structure of skin of Tianzhu white yak,Wnt10 b,FGF18 gene in skin hair follicles.The study on the expression of transcription and protein levels revealed the role of Wnt10 b and FGF18 genes in the cyclical development of the white yak hair follicle and its molecular regulation mechanism.The results showed that:1.Tianzhu white yak skin hair follicle periodic changes: In January and March,it was found that the primary hair follicles and secondary hair follicles did not change significantly,and the hair follicles were small in diameter,and there were cavities in the hair follicles,and the hair follicles were resting.In June,the hair follicle cavity was further increased,but the number and diameter of hair follicles were significantly larger than in January and March.The primary hair follicles and secondary hair follicles were clearly visible.In August,the diameter of hair follicles was further increased.Large,hair follicle cavity decreased compared with January,March and June,when the hair follicle development was in the period of regression;in October and December,the hair follicle cavity almost disappeared,the hair follicle was more visible in the visual field,and the hair follicle grew into the growth phase.According to the statistics of hair density and S/P ratio of Tianzhu white yak,the number of hair follicles in each month was in the process of dynamic equilibrium,and there was no difference.2.Real-time fluorescence quantitative results showed that the expression levels of Wnt10 b gene and FGF18 gene were significantly changed from 1 to 3 months and 8 to 10 months,and the opposite trend was observed,indicating that this is the skin follicle cycle of Tianzhu white yak.The time node of sexual development,that is,the growth period to the regression period,the rest period to the growth period node.3.Immunohistochemistry results showed that Wnt10 b and FGF18 were positively expressed in the dermal papilla,inner root sheath,outer root sheath,stroma,and epidermis,but the Wnt10 b gene was positively expressed in October compared with other months,while FGF18 High expression of genes appeared in August.
