Wood decay is the due to wood decay fungi caused by the wood bad and disintegration of the phenomenon.In recent years,the phenomenon of wood decay is very common in China.Wood decay causes great economic losses to the forestry production.Wood and human relations are very close,human life and production can not be separated from the use of wood.However,with the rapid development of C hina's economy,there is a serious shortage of forest resources in china.In order to improve the quality of wood and reduce wood decay caused by the phenomenon of wood damage.The identification and research of wood decay fungi is particularly necessary.Cellulose is a kind of renewable resource.The degradation of wood cell wall mainly includes the action of cellulase and ligninase.The activity of cellulase of wood decay fungi is determined and the optimum conditions of enzyme production are found.For the transformation of pathogenic woody decay fungi to the beneficial direction for the biological paper and wood processing industries to provide pre-production basis.In this thesis,the fruit bodies of wood decay fungi were collected from the live trees of the forest area.The strains were isolated and purified by the separation and culture medium.Two strains with good viability were obtained and the woody decay type was distinguished by the method of guaiacol.The genomic DNA was extracted and the DNA sequence was amplified by molecular biology based on PCR.The amplified products were purified and sequenced.Seven pairs of specific primers and phylogenetic tree were designed and analyzed.The two strains of wood rot fungi were identified on the molecular level.The res ults showed that the decay fungi Y1 is Fomitopsis pinicola and NCBI landing number is KM373236.1.The wood decay fungi Y2 is Fomes fomentarius and NCBI landing number is HM58410.1.At the same time combined with the traditional morphological method identification two fruiting fungi.Observe and record the morphology characteristic,growth rate,colony color,mycelial odor and mycelial structure changes of the colony's new area.The results of the two methods were compared,and the results of the two tests were basically the same.However,the traditional morphological identification method is based on past experience and reference books on the observation and identification of bacteria.O nce the fruiting body structure changes,the method of identification is not accurate.The wood decay fungi colony culture cycle is longer and the identification time is relatively long.So the use of molecular biology method can be more rapid and accurate identification of wood decay fungi.Can be combined with two identification methods,can be more reliable identification of wood decay fungi.In this paper,the activity of cellulase was analyzed on the basis of identification of wood rot fungi.The fermentation experiment showed that at the initial pH 6.0,lactose 15g/L,KH2PO4 0.5g/L,ammonium nitrate 15g/L,MgSO4 0.5g/L,VB1 0.4g/L,the amount of liquid is 80 mL for 5 days the cellulase activity was the highest.The medium was glucanase(CMCase)was 116.94U/mL,the experiment provides a preliminary basis for the further study of the mechanism of brown rot fungi to degrade cel ulose.
