In recent years,the forest resources in our country is facing a serious problem of shortage,the contradiction between supply and demand of wood is increasingly sharp.How to use and save the wood resources is a problem which is in badly need of solution.During the factors affecting the quality of wood,wood decay caused by wood-decaying fungi is very common. Wood decay kills the lose of a large number of wood resources,reduces the quality of the wood and it seriously affect the use of wood.In order to protect the timber resources and take fully of wood,people have done extensive research on wood decay and wood-decaying fungi.At present,there are two ways to identify wood-decaying fungi,one is morphological identification,witch means through observing the macro and micro cultivation of the colony to identify wood-decaying;The other one is molecular biology method developed in recent years,which means identify wood-decaying fungi rapidly by PCR.Firstly,we make a good deal of experiments to explore the optimum growth tempOerature,humidity and illumination intensity of two kinds of wood-decaying fungi. Then through morphological identification,we observe and record colonies’ growth rate,shape,texture,color and the color change of medium,the mycelium characteristics on the edge of colony etc. Identify wood-decaying fungi by morphological identification. Finally come to a conclusion:wood-decaying fungi A belongs to Polyporaceae,Trametes; wood-decaying fungi B belongs to Polyporaceae,Pycnoprus.Then,we design a couple of primer, optimize the PCR conditions,refine and amplify DNA,do agarose gel electrophoresis,,sequencing,and compare the sequence with BLAST database by Genebank. Then come to a conclusion:wood-decaying fungi A is Trametes versicolor (L.) Lloyd; wood-decaying fungi A belongs to Polyporaceae,Trametes.During research,we discovered:in the identification of strains, along in the use of conventional identification methods was difficult,and prone to errors,the combination of molecular identification techniques can be applied to the identification of strains is more convenient,more accurate results. Molecular identification techniques for the identification of wood-decaying fungi is on the basis of the molecular level,but there are questions should be combining with the other techniques such as morphological character,is easier to identify disputed strains on line.
