Chinese medicinal material of Guangcigu is the dried bulb of Tulipa edulis(Miq.)Baker,which is belonged to Liliaceae family.In recent years,the demand for Guangcigu was increasing,because it has been used for treatment of breast cancer and gout.The source mainly comes from wild herbs presently,but it was decreasing and behind the requirement,because of the widely used of herbicide as well as unreasonable collection.The only way to solve the conflict between demand and supply was to explore the techniques in artificial cultivation and finally achieve standardized cultivation.There were still many problems unsolved to achieve this goal:Endodormancy of bulbs spend a long time which caused longer production cycle and low land utilization ratio.In the natural habitat,the bulbs began to sprout after dormancy released.but it had a long emergence time for mostly forty days and uneven emergence uniformity which was not convenient for cultivation management.The bulb yield was low in the natural habitat.In the later period of growth,sprout tumble appeared easily as a result of the increasing temperature and sunshine which was not good for raising bulb yield.To provide a basis for cultivation,the effect of low storage temperature on dormancy and the influence of salicylic acid treatment on sprout,bulb development and sprout tumble in T.edulis were studied.The main conclusions were as following:(1)The results indicated that storage at 4? for 10 weeks could release bulb dormancy and 6?8 weeks of cold storage was deemed to be the key period of dormancy breaking preliminary.Storage at 4? could prolong growth period and improve the yield of bulb.(2)Above-mentioned research affirmed that storage at 4? could release bulb dormancy of T.edulis and on this basis that we expand the range of temperature and cultivation tests after different storage time was added.Results indicated that varying storage degrees at 4?15? could release bulb dormancy,higher temperature(10?.15?)was good for rooting and bud elongation and lower temperature(FT,4?)was good for emergence.It could be the sign of dormancy release when the bud top came out of outer scales.The results indicated that storage at FT for 8 weeks and 4? for 10weeks were the best treatments for the dormancy release which favored emergence and emergence uniformity.(3)Different concentrations of SA treatment had different effects on growth and development of T.edulis.Among the treatments,10 mmol·L-1 SA treatment was of the most efficiency for emergence and rooting.0.1 mmol·L-1 SA treatment could promote fresh weight and diameter of bulbs.10 mmol·L-1 SA spraying treatment promoted fresh weight and dry weight of bulbs at havest.Spraying a certain concentration of SA could improve Chl content,Pn and Tr,but inhibit Gs and Ci,which was good for the photosynthesis in the period of sprout tumble.It also improved the content of Pro and total soluble protein,inhibited accumulation of MDA and kept SOD activity in a high level.SA treatment was benefit to enhance heat resistance of T.edulis and delay sprout tumble.
