Five Northeast Zi geese 8-month-old were selected in this experiment and their follicular granulosa cells were extracted and primary cultured to study the effect of Alpha enolase(ENO1)related to laying performance genes of Zi geese on the follicular development.The primary granulosa cells were divided into the control group ? the Ad-CMV-Null and Ad-CMV-ENO1 groups.Total RNA of follicular granulosa cells Zi geese were extracted with Trizol method,reverse-transcribed into cDNA,and then amplified with the PCR to obtain traget fragment.When ENO1 overexpressed in the primary cultured granulose cell,the expression levels of Bcl-2 mRNA and Caspase-3 mRNA were detected with real time quantitative RT-PCR.After the application of molecular biological information analysis software,the results were as follows:1.ENO1 mRNA and protein levels of ENO1 overexpression group were significantly higher than that of adenovirus vector group and medium group(P<0.01)by real time RT-PCR and western blot,and when MOI=350,transfection 48 h,and the transfection was 100%.The results show that the constructed ENO1 expression vector can be overexpressed efficiently in follicular granulosa cells of Zi geese.2.As ENO1 overexpressed,the expression levels of Bcl-2 mRNA,related to apoptosis of follicular granulosa cells of Zi Geese,were significantly increased(P<0.01),but the expression levels of Caspase-3 mRNA were significantly decreased(P<0.05).The results showed that ENO1 could regulate Bcl-2 and Caspase-3 related to apoptosis genes.to improve the follicular development of Zi geese by promoting division and inhibiting the apoptosis of follicular granulosa cells of Zi geese.The research plays the key role in studying ENO1 function and mechanism of poultry follicular development.
