| Objective:This topic through animal experiment, to observe the mechanism of action of huanglian jiedu decoction to promote gastric ulcer healing and provide experimental basis for his own ulcers toxic heat syndromes.Method:Will 72 healthy SD rats, male and female half, randomly divided into six groups, namely blank group, model group, huanglian jiedu decoction high, medium and low dose group, each group of 12. Except the blank group, the rest of the group to be pepper spray+ethanol mixed suspension preparation gastric lavage thermal model, then use the acetic acid injection made gastric ulcer model in rats, after composite model built successfully, the blank group and model group give saline lavage, Chinese traditional medicine group of huanglian jiedu decoction high, medium and low dose lavage, ranitidine group of western medicine to fill the stomach,1/d, for two weeks in a row. Rats lavage after dosing, put to death, for femoral artery blood, using Elisa method to detect the contents of SOD and MDA in serum, open abdomen to take out the full stomach, cut open along the greater curvature stomach, ulcer area is measured with vernier caliper, take ulcer scar tissue section, HE staining and observed in the rat gastric mucosal tissue. The data obtained by SPSS 17.0 statistical processing.Results:(1) The serum SOD content:①Compared with model group and the ranitidine group and huanglian jiedu decoction dose group can different degree of rat serum SOD content, with significant difference (P<0.01); ②Ranitidine and group comparison, huanglian jiedu decoction, low dose group of ranitidine rats serum SOD levels lower than group, with statistical significance (P< 0.05); Ranitidine huanglian jiedu decoction high dose group and group there was no significant difference (P> 0.05);③Compared with huanglian jiedu decoction high dose group, huanglian jiedu decoction, low dose group of rats serum SOD content is lower than that of huanglianjiedu decoction high dose group, with statistical significance (P<0.05); Between huanglian jiedu decoction, low dose group had no significant difference (P>0.05).(2) Serum MDA content:①Compared with the model group and the ranitidine group and huanglian jiedu decoction dose groups all can reduce rats serum MDA content in different degrees, with significant difference (P< 0.01);(2)Ranitidine and group comparison, huanglian jiedu decoction, low dose group of ranitidine rats serum MDA content was higher than group, with statistical significance (P< 0.05); Ranitidine huanglian jiedu decoction high dose group and group there was no significant difference (P>0.05);③Compared with huanglian jiedu decoction high dose group, huanglian jiedu decoction, low dose group of rats serum MDA content is higher than that of huanglian jiedu decoction high dose group, with statistical significance (P< 0.05); Huanglian jiedu decoction, low dose group had no significant difference (P> 0.05).Conclusion:Huanglian jiedu decoction can significantly reduce rat gastric mucosal tissue pathological grading, promote ulcer healing and its mechanism of action may by increasing the rat serum SOD content and decrease MDA level; Huanglian jiedu decoction of ranitidine high dose group and treatment group were superior to middle and low dose group, at the same time within a certain range are dependent of drug concentration. |
