Objective:This study takes rats which were induced to acute gastric mucosal lesion by absolute ethanol as subjects to study the influence of electroacupuncture pretreatment and moxibustion pretreatment at Zusanli and Zhongwan on proliferation and repairrelative facts such as EGF, TGF-? and PCNA of acute gastric mucosal lesion rats, to discuss the difference of the two effective interventions for promotion, proliferation, repair and reconstructionthe of gastric mucosal cells. Methods: 40 SPF-class SD rats were randomly allocated to four groups: Normal Group, Model Group, Electroacupuncture preconditioning(EA) Group and Moxibustion preconditioning Group. After 8 days' pretreatment of moxibustion or electroacupuncture at Zusanli and Zhongwan according to different groups, we adopt absolute ethanol gavage( 0.5ml/100g)to create Acute Gastric Mucosal Lesion models. One hour after creating the models, the rats were anesthetized through intraperitoneal injection by 20% urethane(0.6ml/100g), and then we fetch gastric tissue. Visually observe the gastric mucosal injury and record the injury-index. Observe the histomorphological changes of the gastric mucosa under light microscope. Detect the content of EGF in serum by enzyme immunoassay. Detect the expression of TGF-? and PCNA in gastric mucosa by immunohistochemistry method. Results: 1. Compared with Normal Group, gastric mucosa of Model Group was obviously injured and the injury-index was significantly higher(p< 0.01), the histomorphological structure was markedly destroyed under light microscopic. Compared with Model Group, the gastric mucosa damage degree of Electroacupuncture Pretreatment Group and Moxibustion Pretreatment Group were lower, the injury-index were significantly lower( p<0.05 or p<0.01), and the histomorphological structure were more complete. There was no significant difference between Electroacupuncture Pretreatment Group and Moxibustion Pretreatment Group.2. Compared with Normal Group, the content of EGF in serum of Model Group was significantly lower(p<0.05), and the expression of TGF-? and PCNA in gastric mucosa of Model Group were significantly higher(p<0.05). Compared with Model Group, the content of EGF in serum and the expression of TGF-? and PCNA in gastric mucosa of Electroacupuncture Pretreatment Group and Moxibustion Pretreatment Group were higher in different degrees(p<0.05 or p<0.01). Compared with Electroacupuncture Pretreatment Group, the expression of TGF-? in gastric mucosa of Moxibustion Preconditioning Group was statistically higher(p<0.01), but there was no significant difference of the content of EGF in serum and the expression of PCNA in gastric mucosa between the two groups. Conclusion: EA pretreatment and Moxibustion pretreatment were protective for acute gastric mucosal lesion, both of them can reduce the damage on gastric mucosa which was induced by absolute ethanol, and the effect of moxibustion pretreatment was superior to that of electroacupuncture pretreatment. The mechanism may be related to gastric mucosal protective factors(EGF, TGF-?,PCNA)...
