Objective:Through detecting CAIX and PTEN protein expression in renal cellcarcinoma and normal kidney tissue, I makeanalysis of the relation-ship between theexpression strength of CAIX and PTEN protein expression in renal cell carcinomaandpathological grade, clinical staging, recurrence and lymph node metastasis, and make the Pearson rank correlation analysis. Explore the relationship of both CAIXand PTEN protein and the development of renal cell carcinoma as well as its value indiagnosis and treatment.Methods:Collecting paraffin specimens from 98 cases of renal cell carcinoma and normal kidney specimens from 10 cases, in which the patients were all from the First Affiliated Hospital of Bengbu Medical College from January 2008 to August 2011. We adopted the method of immunohistochemistry, SP, to assay CAIX and PTEN proteinexpressed in the tissues and to analyze its significance.The data was analyzed bySPSS13.0 statistical software.Results:(1)CAIX expression in clear cell renal carcinoma tissues and normal renal tissues.The postivite expression rates of CAIX in clear cell renal carcinoma tissues andnormal renal tissues are 72.45%(71/98) and 0.00%(0/98) respectively.There is a significant difference between the expression of CAIX in the two kinds of tissues.(χ2=29.2384, p). The expression of CAIX has no relation to the difference in the age of patients,the diameter of the tumors,TNM staging,Fuhrman classification or whether there is lymphatic metastasis.(2)PTEN protein expression in clear cell renal carcinoma tissues and normal renal tissues.The postivite expression rate of PTEN protein in clear cell renal carcinoma tissues and normal renal tissues are 42.86%(42/98) and 100.00%(98/98) respectively.There is significant difference between the expression of PTEN protein in the two kinds of tissues. (χ2=16.9925, p). The expression of PTEN protein has no relation to the difference in the age of patients,the diameter of the tumors,but thepositive expression rate in patients with lymphatic metastasis was significantly lower than that in patients without lymphatic metastasis.In the histolological classication of Fuhrman,the positive expression rate of Ⅰ、 Ⅱ and Ⅲ~Ⅳ stage were 71.19%、31.36%、0.00% respectively, showing a decreasing trend (r=-0.387, p). In clinical stage, the positive expression rate of Ⅰ、 Ⅱ and Ⅲ~Ⅳ stage were were 81.82%、53.19%% 5.56% respectively, showing a decreasing trend (r=-0.314, p) at the same time.Conclusion:1、The expression of CAIX in clear cell renal carcinoma tissues was up-regulated compared to the nomal renal tissues,which has no relation to the difference in the age of patients,the diameter of the tumors,TNM staging,Fuhrman classification or whether there is lymphatic metastasis. The expression of PTEN protein in clear cell renal carcinoma tissues was inhibited compared to the nomal renal tissues,which has no relation to the difference in the age of patients and the diameter of the tumors,but was negatively correlated with the TNM staging and Fuhrman classification.What’s more, the positive expression rate in patients with lymphatic metastasis was significantly lower than that in patients without lymphatic metastasis.From what has been disscussed above,we can conclude that CAIX contributes to the diagnosis of clear cell renal carcinoma and the expression of PTEN protein has relation to the clinical and pathological characteristics of clear cell renal carcinoma. The detection of CAIX and PTEN protein is helpful to assessement of the cell carcinoma’s severity and its prognosis.2、The expression of CAIX in clear cell renal carcinoma tissues was up-regulated compared to the nomal renal tissues and the expression of PTEN protein in clear cell renal carcinoma tissues was inhibited compared to the nomal renal tissues,proving that both are closely to the occurrence and development of clear cell renal carcinoma.The co-expression of CAIX and PTEN protein in clear cell renal carcinoma can predict the recurrence and prognosis of the renal cell carcinoma,can be used as a new marker of clear cell renal carcinoma, also providing a new way for the early diagnosis of renal cell carcinoma. CAIX and PTEN protein in clear cell renal carcinoma may become a new target of gene theraphy,offering new ideas and research directions in gene level for the treatment of clear cell renal carcinoma.
