Objective In this study,we observation the Heparin on burn wound healing in the case of nano-silver antibacterial gel control rat skin deep ?°burn wound infection.With the wound tissue expression of PCNA,KGF and the changes of collagen fibers,to determine whether the impact wound healing cell proliferation,keratinocytes forming cells,the formation of collagen fibers,and to explore the physiological mechanism of pathology heparin on burn wound healing,provide a theoretical basis for clinical treatment.Methods 66 SD rats,weighing 190±10g,regular feeded at a temperature of 20 ~ 25 ?,humidity around 50% of the room after the UV disinfection.They were randomly divided into normal control group,model determination group,model group,sterile dressing group,nanosilver group,heparin group and the combined drugs group,all the rats' hair on the backs near the tail side was removed,the last 6 groups' hair removal used the method of closing to the water continued boiling flask mouth(diameter 2cm)5S production burn model.Model rats exposed burn wounds,sterile dressing groups,nanosilver group,heparin group and the combination group rats were burn wound with sterile gauze,rats nanosilver group,heparin group and combination group were burn wounds with heparin,nano-silver antibacterial gel and 1ml both applied to the wound.Rats were observed daily mental state,weight,wound situation,when drawn up,the normal control group,model group were drawn after molding 1d,model group,sterile dressing group,heparin,nano-silver group and combination group were in the first 4,14,21 d and healing drawn,every time each animal were drawn 3,Anesthetized rats after burn wound along the edge of extended 5 mm cut tissue thickness and full-thickness skin deep muscle partial.Formalin-fixed,paraffin sections,the surface which was embedded with a burn trauma center edge,cut into thick 6-7 ?m.After HE,Sirius red,PCNA and KGF immunohistochemical staining,microscopy,semi-quantitative and statistical analysis using collagen volume fraction,PCNA and KGF positive cells.Results1.The determination of deep ?°burn Model Thinning of the skin area burns,skin necrosis,an island residue;damage the epidermis,dermis and dermal part of the skin appendages,which suggest that the deep II°degree burn of rat model was successfully established.2.The impact of heparin nano-silver antimicrobial gel for deep?°burned rats general situation burned rats 100% survival,spiritual,diet and urine and found no abnormalities,weight increased.After injury 14 d,heparin group and nano silver body weight than the model group(P<0.05),the combined group was significantly heavier than the model group,sterile dressing groups,nanosilver group and heparin group(P<0.01),and until healed.Tip heparin nanosilver may relieve local discomfort burns better combination effect.3.Impact of heparin nano-silver antimicrobial gel on Deep ?°Burn Wound rats deep ?°burn wound infections were not found,and gradually wound healing,healing time is substantially different in each group,the combination group healing first,followed by the heparin group,followed by nano-silver group and sterile dressing,model the latest healing time(P<0.05).4.The histopathology effect of heparin in combination with nano silver antibacterial gel on burn wounds in rats Rats wound in each group over time,leakage reduction,skin and its appendages,dermal are repaired.When combined group 4d mild inflammation,repair the structure close to the normal control group at 21 d,while healing skin appendages appear.5.The effect of heparin in combination with nano silver antibacterial gel on rat burn wound collagen fibers Sirius red staining of collagen fibers appear red.When 4d,the amount of collagen fibers combined treatment group was lower than model group,sterile dressing groups,nanosilver group and heparin group(P<0.05).When 14 d,the combination group than the model group,sterile dressing groups,nanosilver group(P<0.05),and no difference in the heparin group;heparin group compared with the model group(P<0.05).At 21 d and healing,no significant difference between the groups of collagen.In the healing process,collagen fibers of a decreasing tendency with the prolongation of time after injury,the amount of change of collagen fibers in ascending order: the combined group,heparin,nano-silver group,sterile dressing,model.6.The impact of heparin in combination with nano silver antibacterial gel for burns skin KGF on rats KGF positive in the cytoplasm,brownish yellow fine granules.KGF expression in model group at 4d low expression increased at 14 d,but lower than the control group(P<0.01),when the healing is close to the normal control group.When 4d,combined treatment group than the model group,sterile dressing groups,nanosilver group and heparin,heparin group than in the model group(P<0.01).When 14 d,the combined group,heparin group and nano-silver group than the model group(P<0.05).At 21 d and healing,each group of similar,close to the normal control group.7.The effect of heparin in combination with nano silver antibacterial gel on rat burn wound cell proliferation PCNA-positive nuclei located,was brown.PCNA positive rate of epidermal cell model in 4d group than normal control group(P<0.05),14 d higher than the normal control group and continued to 21d(P<0.05),close to the normal control group at the time of healing.When 4d,the combined group,heparin,nano-silver group,sterile dressing with the model group were similar.When 14 d,combination therapy group was significantly higher than the model group,sterile dressing groups,nanosilver group(P<0.01),and no difference in the heparin group;sterile dressing group,nano-silver group and heparin group than the model group(P < 0.05).When 21 d,the combined group than the model group,sterile dressing groups,nanosilver group(P<0.05),and no difference in the heparin group;nano-silver group,heparin group than in the model group(P<0.05),sterile dressing group and model group no significant difference.When healing,each group of similar,close to the normal control group.Dermal connective tissue of PCNA positive rate of model group 4d,14 d was significantly lower than the control group(P<0.01),in the 21 d,when healing close to the normal control group.When 4d,the combined group,heparin group,and a sterile dressing nanosilver group and model group were similar.When 14 d,combination group and heparin group than the model group(P<0.05).When 21 d,the combined group than the model group,sterile dressing groups,nanosilver group,heparin group(P<0.05);at the time of healing,the model group than the control group(P<0.05),sterile dressing group nanosilver group,heparin group,combination therapy group than the model group,wherein the heparin group and the combination group was significantly higher than the model group(P<0.05).Conclusions1.Heparin in combination with nano silver antibacterial gel on rat skin deep ?°burn wound healing superior to heparin or nano-silver,and heparin were better effective than nano silver.2.Heparin,nano-silver antibacterial gel on rat skin deep ?°burn wound healing,witch may through slow the formation of collagen fibers wound,promote the formation and differentiation of keratinocytes and wound induced cell proliferation.
