Objective:To detect the expression of Stathmin protein and STAT3 protein in gallbladder carcinoma(GBC),Gallbladder adenoma and chronic cholecystitis tissues.Investigating the relationship between those two proteins and clinicopathological parameters of gallbladder carcinoma,and explore the role of Stathmin protein and STAT3 protein in the carcinogenesis and development of GBC.Methods:The paraffim specimens were collected from the First Affiliated Hospital of Nanchang University,including 25 cases of GBC tissues,15 cases of Gallbladder adenoma tissues and 15 cases of chronic cholecystitis tissues.All patients didn,t undergo radiotherapy,chemotherapy and other adjuvant therapy before surgery.The expression of those two proteins were detected by using SP immunohistoch emistry(IHC),then the experimental data were statistically analyzed by using SPSS20.0,and investigating the relationship between the positive expression of two proteins and the clinicopatholog-ical parameters of gallbladder carcinoma.Results:1.The positive expression rates of Stathmin protein in GBC tissues,gallbladder adenoma tissues and chronic cholecystitis were 72%,33.3% and 26.7%,respectively;the positive expression rate of Stathmin protein in GBC was higher than that in nongallbladder cancer tissues and the difference was statistically significant(P <0.05);the expression of Stathmin protein was related to the differentiation,clinical stage and lymph node metastasis of gallbladder carcinoma,while it has nothing to do with the patient's age and sex(P>0.05).2.The positive expression rate of STAT3 in GBC tissues,gallbladder adenoma tissues and chronic cholecystitis tissues were 60%,20%,26.7%,respectively.The positive expression of STAT3 protein in GBC tissues was significantly higher than that in other tissues,and the difference was statistically significant(P<0.05).the expression of Stathmin protein was related to the clinical stage and lymph node metastasis of gallbladder carcinoma,while it has nothing to do with the patient's age and sex(P>0.05);while,there was no significant difference in the expression of STAT3 in the high,medium and low differentiated gallbladder carcinoma tissues,But the positive expression rate of the 3 groups increased with the decrease of the degree of differentiation.3.The expression of Stathmin and STAT3 in GBC tissue was positively correlated.Conclusions:1.Stathmin and STAT3 protein did not express or low express in chronic cholecystitis and gallbladder adenoma tissues,but were highly expression in GBC tissues,indicating that Stathmin and STAT3 may be involved in the occurrence and development of GBC.2.The expression of Stathmin and STAT3 protein were related to the differentiation,clinical stage and lymph node metastasis of gallbladder carcinoma,while it has nothing to do with the patient's age and sex.3.It is highly expression of Stathmin and STAT3 protein in GBC tissues,suggesting that they may have certain correlation and the interaction between the two proteins may be involved in the occurrence and metastasis of gallbladder carcinoma.Cooperative detection of those two proteins may contribute to the invasiveness and prognosis of GBC.In addition,those two proteins are expected to become the target of the targeted therapy of gallbladder cancer.
