Objective: To study the bone marrow stroma stem cells mobilization effect of trichosanthes kirilowii Maxim on the rat model of hyperlipidemia combined with myocardial ischemia syndrome and the antioxidation effect. As the entry point to the mobilization effect on bone marrow stroma stem cells, It reveals the mechanism of the medicine intervene the hyperlipidemia combined with myocardial ischemia syndrome.Methods:(1) Male Wistar rats were randomly divided into five groups, which are the normal control group, the simple myocardial ischemia group, the hyperlipidemia control group, the syndrome model group and the trichosanthes kirilowii Maxim intervention group. To get the hyperlipoidemia model, we make the rats treate with high-fat emulsion. Then, left anterior descending coronary artery was ligatured to simulate the acute myocardial ischemia model. A sham-operation was implemented on the normal control group. The trichosanthes kirilowii Maxim intervention group were given the trichosanthes kirilowii Maxim decoction(20 g/kg) treatment on the basis of the model. Then we used the semi-automatic biochemical analyzer and biochemical assay kits to detect the plasma content of superoxide dismutase(SOD)?glutathion Peroxidase(GSH-P X)?catalase(CAT)?total antioxidant capacity(T-AOC)?Nox?lipid peroxide(LPO)?oxidized low density lipoprotein(ox-LDL)in each group.(2)Bone marrow stroma stem cells mobilization detection:Another male wistar rats were divided into five groups, which were the normal control group, the simple myocardial ischemia group, the hyperlipidemia control group, the syndrome model group and the trichosanthes kirilowii Maxim intervention group. Those groups were builded methods as fore-mentioned.These rats were randomly divided into 7 groups, namely: the first to seventh subgroup. In 1-7 days, the rats were sacrificed after coronary artery ligation respectively, stained by the CD34(PE)?v WF(FITC)antibodies and taken peripheral blood to detect the bone marrow stroma stem cells level in peripheral blood by flow cytometry, then the dynamic curve was drawn.Results:(1)When compared with the normal control group, the SOD?T-AOC?CAT values of the syndrome model group were significantly reduced while the LPO?GSH-P values were significantly increased.When compared with the syndrome model group,the SOD?T-AOC?CAT?GSH-PX values of the trichosanthes kirilowii Maxim intervention group were significantly increased and they were exceeded than the normal control group,the LPO values of the trichosanthes kirilowii Maxim intervention group were significantly decreased and they were lower than the normal control group.When compared with the hyperlipidemia control group,the ox-LDLvalues of the syndrome model group had no obvious difference,but the NADPH values were increased.When compared with the syndrome model group,the ox-LDL?NADPH values were decreased,and they were lower than the hyperlipidemia control group(P<0.05 or P<0.01).(2)Flow cytometry datas showed that peripheral blood content of bone marrow stromal stem cells of the normal control group and the hyperlipidemia control group has been in a low level and keeps no change.Peripheral blood content of the simple myocardial ischemia group increased in 1 to 6 days, in 6 days to peak, the seventh day down to a lower level,but still higher than that of normal control group.The variation trend of the simple myocardial ischemia group and the syndrome model group keeps consistent, each group overall level is lower than the simple myocardial ischemia group(P<0.05 or P<0.01).Com- pared with the syndrome model group,the trichosanthes kirilowii Maxim intervention group presented upward tendency and reached peak on 7st day(P<0.05 or P<0.01).Conclusion:Myocardial ischemia is intense stimulation factors of bone marrow stromal stem cells mobilization,as one of myocardial ischemia induced factors of hyperlipidemia,it can lead to bone marrow stromal stem cells mobilization. Snakegourd fruit skin water decoction can significantly reduce the rats blood lipid of hyperlipidemia complicating acute myocardial ischemia and improve the activity of antioxidant enzymes to protect the damaged myocardium.It can aslo significantly improve the rat bone marrow stromal stem cells mobilization of hyperlipidemia disease complicating acute myocardial ischemia.
