Objective This topic uses healthy male KM mice,CTX was used by abdominal cavity injection once a day for three times to duplicate CTX-induced models with myelosuppression. By observing the regulation of acupuncture and moxibustion on marrow ICAM-1,VCAM-1 expression in post chemotherapy mice, to investigate the influence of acupuncture and moxibustion on marrow hematopoietic microenvironment, then from the angle of which, to explain the mechanism of acupuncture and moxibustion on improving marrow supression after chemical therapy, promoting recovery of hematopoietic function and enhancing leukocytes in peripheral blood.Methods Select seven weeks old healthy male KM mice, according to body weight, then randomly divide into normal group,model group, acupuncture group and moxa group. The normal group was injected with 0.9% NS according to same volume. The same doses of CTX was used by abdominal cavity injection (According to doses of 0.02ml/g for per mouse with the concentration of 5mg/ml) for the rest three groups of mice,followed by repeated injection 24h and 48h afterwards continuously for 3 days, the models of myelosuppression were estabished 4h post repeated injections. Select points of"Da Zhui","Ge Shu ","Shen Shu"and"Zu San li", then continuously apply acupuncture and moxa to the mice 4h post duplication in the corresponding groups once daily,normal group,model group accompanied fixed. In addition, before duplication and 3 days, 5 days afterwards, cutting the tails to obtain blood and detect the leukocytes counting in peripheral blood respectively, then kill a batch of mice and take out thighbone, fix the sample with 4% polysorbate, embedded by paraffin wax, slice it into 4μm thick. Observe the expression of adhesion molecules as ICAM-1,VCAM-1 with Immunohistochemical method. Statistical software of SPSS13.0 was used for data analysis.Results1.Acupuncture and moxibustion can elevate WBC counts in post chemotherapy mice. White blood cell in peripheral blood after molding dropped in the model, acupuncture and moxa groups ,and the difference(P<0.05)was obvious compared with the normal group; while,three days after therapy, white blood cell in peripheral blood in the acupuncture and moxa groups rised compared with the model group, the difference of which(P>0.05)was not obvious. the acupuncture group increased obviously (P>0.05) than the moxibustion group; Five days after treatment, WBC counts of acupuncture, moxa groups rised continued to rise, the difference between the acupuncture group and the control, model groups were obvious (P<0.05),WBC counts rised also in the acupuncture and moxibustion groups, but there was not significant difference(P>0.05).2.Three days after therapy, the expression of adhesion molecule ICAM-1 increased in the acupuncture and moxa groups, with remarkable difference(P<0.05)compared with the normal group. The ICAM-1 expression in the moxa group was higher than the acupuncture group, and there was not significant differenc(eP>0.05); Five days after treatment, The ICAM-1 expression in the acupuncture and moxa groups both rised significantly(P<0.05) compared with the model group, the level in the moxa group was higher than the acupuncture group while without remarkable difference(P>0.05).3. Three days after therapy,compared with the model group, the expression of adhesion molecule VCAM-1 in mice bone marrow were both increased in the acupuncture and moxa groups without significant difference(P>0.05), and the level in the moxa group was higher than the acupuncture group without obvious difference(P>0.05);Five days after treatment, the VCAM-1 expression in the acupuncture and moxa groups were both increased, while without significant difference(P>0.05)compared with the normal group,and the level in the moxa group was higher than the acupuncture group, but without obvious difference(P>0.05).Conclusions1. Acupuncture and moxibustion can definitely relieve myelosuppression in mice post chemotherapy with CTX, promote recovery of hematopoietic function and elevate the leukocytes in peripheral blood, the effects increase along with the times of treatment; Based on tutor's previous researches, acupuncture and moxibustion can elevate WBC counts, and the effect was exact.2. CTX can downgrade the expression of marrow ICAM-1, acupuncture and moxibustion can promote marrow expression of ICAM-1 in CTX-induced mice, benefit the adhesion and information exchange between hematopoietic cells and tissues, enhance sensibility of hematopoietic cells to cytokine, regulate hematopoietic function, help repair marrow injury of hematopoietic microenvironment, and then elevate the leukocytes in peripheral blood.3. CTX can downgrade the expression of marrow VCAM-1, acupuncture and moxibustion can promote marrow expression of VCAM-1 in CTX-induced mice, promote the integration of hematopoietic cells and stromal cells, which help proliferation and differentiation of hematopoietic cells, and then elevate the leukocytes in peripheral blood.
