Objective: To investigate the expression level of telomere associated protein dyskerin,MRE11, Ku80 gene and the related syndrome differentiation of TCM of Chronic Aplastic Anemia(CAA), and to study the pathogenesis of Aplastic Anemia.Methods: Extracted bone marrow from both CAA patients and normal patients, using real time fluorenscene quantitative PCR technology, to detect the mononuclear cells telomere associated proteins dyskerin, MRE11, Ku80 gene, expression level of m RNA in bone marrow, to explore dyskerin, MRE11, Ku80 gene and expression level of mRNA and relationship between the control group, to analyze the differences of gene expression between Aplastic Anemia patients with different syndrome differentiation.Results: 1. Dyskerin gene: comparison between control group and CAA gorup, there is differences within dyskerin gene mRNA expression level(P<0.05), there is statistical significance, the expression level of dyskerin gene in CAA group is lowered; Kidney-Yin deficiency group, Kieney-Yin and-Yang deficiency group and Kidney-Yang deficiency three groups compare with the control group. The former two genes expression level are lowered(P<0,05), CAA with TCM syndrome differentiation group comaprison(P>0.05),does not havr statistical significance, there is no differences between the groups, there is no significant differences in the expression of gene.2. MRE11 gene: comparison betwen control group and CAA group, there is comparison within MRE11 gene m RNA expression level(P<0.05), there is statistical significance, and the MRE11 gene expression level is lowered in CAA group, Kidney-Yin deficiency, Kidney-Yin and-Yang deficiency group and Kidney-Yang deficiency three groups compare with the control group, the expression level of the former two groups arelowered, Kidney-Yang deficiency group(P>0,05) compare with the control group there is no significance differences; comparison between chonic AA TCM syndrome differentiation group, compariosn between Kidney-Yin deficiency and Kidney-Yin and-Yang deficiency,its gene expression is lowered(P<0,05), and the gene expression level in Kidney-Yin and-Yang is lowered.3. Ku80 gene: Comparison within control group and CAA group, there is no difference in Ku90 gene mRNA expression level(P>0.05), there is no significant differences meaning in Ku80 gene mRNA expression level in comparing CAA group and control group; Kidney-Yin deficiency group, Kidney-Yin and-Yang deficiency group and Kidney-Yang deficiency 3 groups compare with control group, the expression level of gene of the former two groups are lowered comapre to control group(P<0.05), while there is significant lowered in the expression level in Kidney-Yin and-Yang group; comparison in CAA TCM syndrome differentation, Kidney-Yin deficiency group compare with Kidney-Yang group, its gene expression level is lowered(P<0,05), while the expression level is lowered significantly in Kidney-Yin deficiency group.Conclusion: Patients with chronic Aplastic Anemia with telomere associated protein dyskerin, MRE11 gene mRNA expression level is lowered, the expression level of mRNA is lowered, indicating that the stability of telomere structure and decrease of telomerase activity, further increase the pathogenesis of molecular level in CAA, this is perhaps one of the pathogenesis of chronic Aplastic Anemia, from the gene expression level of three related protein, the expression level of three types in Kidney-Yin deficiency syndrome is lowered in different levels, this is regarding the “Yang deficiency is easily to be treated,Yin deficiency is difficult to be regulated’ in TCM theory provides prove for molecular evidence.
