| Lycopodium japonicum, belonging to Lycopodiaceae, was historically used as a traditional folk medicine for the treatment of contusion, strains, and myasthenia. In order to have a systematic research on the whole herb of Lycopodium japonicum purchased from Bozhou, Anhui, the basic ethyl acetate extract of Lycopodium japonicum was studied. Nineteen alkaloids were isolated, including eleven Lycopodine alkaloids: lycoposerramine-M N-oxide(1), acetyllycoposerramine M(2), lycopodine(3), lycoposerramine-M(4), miyoshianine C(5), 12-epilycodoline N-oxide(6), gnidioidine(7), lycoposerramine-K(8), lucidioline(9), 4α-hydroxyanhydrolycodoline(10), flabelline(11); five Lycodine alkaloids: hydroxypropyllycodine(12), lycodine(13), des-N-methyl-α-obscurine(14), α-obscurine(15), des-N-methyl-β- obscurine(16); three Fawcettidane alkaloids: lycoflexine(17), lycoflexine N-oxide(18) and fawcettidine(19). Wherein the compound 1 is a new, compounds 2, 6, 11-12, 16, 18-19 were isolated from this plant for the first time.The medicinal mushroom Ganoderma lucidum belongs to the family of Polyporaceae. We studied the ethanol extract of Ganoderma lucidum which grown in Xishuangbanna, Yunnan. Ten compounds were obtained including seven Ganoderma triterpenoids: ganoderic acid Y(20),antiquol C(21), 7-oxo-ganoderic acid Z3(22), 7-oxo-ganoderic acid Z(23), ganoderic acid B9(24), ganoderic acid C2(25), 3β,7β-dihydroxy-11,15,23-trioxo-lanost-8,16-dien-26-oic acid(26) and three sterols: ergosterol(27), 5α, 8α-epidioxyergosta-6, 22-dien-3β-ol(28), 6,9-epidioxy-ergosta-7, 22-dien-3β-ol(29). Compound 21 was isolated from this genus for the first time.Anti-inflammatory assay of compounds 1-19 was evaluated against LPS-induced NO releases. An in vitro model of LPS-induced RAW264.7 was established. The level of NO in the culture supernatant was tested by Griess Reagent, and activity of compound was evaluated based on the NO inhibition ratio. Miyoshianine C(5) and lycoflexine N-oxide(18) exhibited potent inhibitory activity with the IC50 values were 31.82±3.74 and 40.6940.69±5.78 μmol/L, respectively. |
