Ganoderma lucidum spore oil is using carbon dioxide extraction technique(SFE-CO2) from broken ganoderma lucidum spore lipids extracted active substances, a collection of active ingredients of ganoderma lucidum spores, used in the treatment of high cholesterol, hepatitis b, tumor and other diseases. Ganoderma lucidum spores oil are widely distributed throughout the country, Fujian province is one of the main production of it. In recent years because of ganoderma lucidum spore oil is expensive, false of ganoderma lucidum spores oil spred everywhere. To evaluate the quality of the Ganoderma lucidum spores oil by TLC, HPLC, GC-MS, GC and ICP-MS. It may provide scientific foundations to the authenticity identification, quality control and the further development and utilization of the Ganoderma lucidum spores oil.Chapters1. determination the Ganoderma lucidum spores oil by the TLC Methods: The protein of Ganoderma lucidum spores oil and other vegetable oils were identified by TLC. Results: The TLC method was established to identity the Ganoderma lucidum spores oil and it can effectively used to qualitative identification. Conclusion: TLC can be accurately used to identify the authenticity of the Ganoderma lucidum spores oil, characteristics of EG spots can be used to distinguish the ganoderma lucidum spore oil with ordinary vegetable oil, but not accurate for the merits of the pseudo-doped and quality identification.Chapters2. The content determination of the EG and ergosterol in Ganoderma lucidum spores oil. Determination the content of the EG and ergosterol in Ganoderma lucidum spores oil. Methods: HPLC and GC-MS were used to determinate the EG and ergosterol in the Ganoderma lucidum spores oil. Results: HPLC method was established to determinate the EG and ergosterol in the Ganoderma lucidum spores oil, the method is simple, accurate and reliable. The concentrations of EG between 0.35-35.20μg/mL indicated a good linear. The rate of recovery was 95.06-98.98% and RSD < 0.63%(n=6). The concentrations of ergosterol between 0.62-62.37μg/mL indicated a good linear. The rate of recovery was 94.96-99.12% and RSD < 0.21%(n=9). Conclusion: HPLC and GC-MS can be accurately used to determinate the EG and ergosterol in the Ganoderma lucidum spores oil, which may be provide scientific foundations to the authenticity identification. Compared with the GC-MS, HPLC chromatographic column sample is not easy to de polluted, simple operation, accurate and reliable.Chapters3. Seven kinds of fatty acid in the ganoderma lucidum spores oil content determination. Determination the content of Seven kinds of fatty acid in the ganoderma lucidum spores oil. Methods: GC was used to determinate seven kinds of fatty acids(Myristate, Pentadecane acid, Hexadecanoic acid, 17 alkanes acid, Stearyl acid, Oleic acid and Linoleic acid)in the Ganoderma lucidum spores oil and other vegetable oils. Results: GC method was established to determinate seven kinds of fatty acid in the ganoderma lucidum spores oil, verified the fatty acids in good linear relationship, RSD < 2.59%, the rate of recovery was 95.06-98.98%(n=9). Conclusion: Ganoderma lucidum spores oil and ordinary vegetable oil fatty acid composition effectively, the results show the ordinary vegetable oil fatty acid composition are similar to those of ganoderma lucidum spores oil, unsaturated fatty acid is given priority to with oleic acid and linoleic acid, saturated fatty acid is given priority to with palmitic acid and stearic acid, almost all batches of ganoderma lucidum spores oil fatty acid amount in more than 90%, ordinary vegetable oil fatty acid amount of about 80%, compared with the ganoderma lucidum spores oil content is relatively low.Chapters4. The paper used Inductively Coupled Plasma Mass Spectrometry for the quantitative analysis of 22 elements in the ganoderma lucidum spores oil. Methods: ICP-MS was used to determinate 22 kinds of elements, include Pb、As、Hg、Cd、Cu、Li、Be、Na、Mg、Al、K、V、Cr、Mn、Fe、Co、Ni、Zn、Se、Sr、Cs、Ba. Results: The analysis method, ICP-MS method was established for the simultaneous determination of 22 trace elements in certified, each element of a good linear relationship. Conclusion: The heavy metal content determination of 20 batches of ganoderma lucidum spores oil samples are comply with the standards set by the 2015 edition of China pharmacopoeia, different origin of ganoderma lucidum spores oil heavy metal content difference is not big, from the test results, ganoderma lucidum spore oil samples most abundant sodium content.Chapters5. Ganoderma lucidum spores oil in vitro pharmacological activity screening. Methods: The vitro pharmacological activity screening of the Ganoderma lucidum spores oil and UMGSO. Results: Ganoderma lucidum spore oil and UMGSO dependented inhibition of human leukemia cells, K562, human lung adenocarcinoma cells(A549), human breast cancer cells(MCF-7) and the added value of liver cancer cells(HepG2). Conclusion: Ganoderma lucidum spore oil and UMGSO are dose dependent inhibition of human tumor cells. Result shows that Ganoderma lucidum spore oil is dose dependent inhibition of human leukemia cells, K562, human lung adenocarcinoma cells(A549), human breast cancer cells(MCF-7) and the appreciation of liver cancer cells(HepG2), the IC50 were12.69μg/mL、20.59μg/mL、34.12μg/mL、37.18μg/mL.UMGSO is also dose dependent inhibition of K562,A549, MCF-7 and Hep G2, the IC50 were 15.15μg/m L、16.94 μg/m L、53.01 μg/m L 及40.38μg/m L. Compared with the GMSO, Ganoderma lucidum spore oil itself have stronger antitumor activity, get rid of the fatty acid and the rest of the may have synergy in antitumor effect.
