Objective:To explore the expression of ribosomal protein L6 in prostate cancer and it`s clinical significance. And study its effect on prostate cancer biology behavior in vitro and it`s molecular mechanism.Methods:The expression level of RPL6 in adjacent non-cancerous tissues and prostate cancer tissues was detected by RT-qPCR and western blot technique. The relationship between clinicpathological factors of prostate cancer and RPL6 m RNA expression level was statistically analyzed by SPSS. Western blot and RT-qPCR technique to detect the protein expression and mRNA level of RPL6 in LNCaP and PC3 prostate cancer cells, and its expression level was also detected in RWPE-1 prostate cancer cells. RPL6 shRNA plasmid was constructed and amplificated befor transfected into PC3 cells. Puro was used to screen the transfected PC3 cells and get the stable cells,in which the level of RPL6 was detected by western blot and RT-qPCR technique.The proliferation capability of RPL6 low expressed cells were assessed by CCK8 assay. The capacity of migration and invasion was detected by transwell in RPL6 low expressed cells. Annexin V assay was used to evaluate the poptosis of RPL6 low expressed cells. Nude mouse tumorigenicity assay was used to assess the tumorigenesis ability of RPL6 low expressed PC3 cells. SPSS statistical analysis software was used to the analysis of experimental result.Results:Compared with non-cancerous tissues the RPL6 were expressed at higher level in prostate cancer tissues(P<0.05). The higher expression levels of RPL6 mRNA was significantly correlated with the increased level of PSA, lymph node metastasis,higher Gleason score, vascular invasion and clinical stage(P<0.05), but the RPL6 expression levels has no link between age, urgical margin status and seminal vesicle invasion(P>0.05). Kaplan-Meier survival analysis suggested that patients with high RPL6 mRNA expression BCR-free survival time were shorter than those with lowRPL6 expression. The RPL6 were at a higher expression level in LNCaP and PC3 prostate cancer cells than in RWPE-1 cells. RPL6 shRNA plasmid was constructed and amplificated befor transfected into PC3 cells. Cell biological behavior tests find that low expression of RPL6 in PCa cells reduce cell invasion, proliferation significantly and tumorigenesis ability in nude mice was also discounted, but the apoptosis of PCa cell was increased.Conclusions:Compared with non-cancerous tissues the mRNA and protein of RPL6 were expressed at higher levels in prostate cancer tissues. The abnormal express of RPL6 in PCa cells have an impact on its biological behavior in vitro and vivo. The higher expression levels of RPL6 were involved in the occurrence and development of prostate cancer and indcated an unfavorable prognostic significance; it can be used as a markerbe to assess the progression and prognosis of prostate cancer.
