To investigate the intervention effect and mechanism of JIANPI RUANGAN DECOCTION on CCL4 induced hepatic sinusoidal endothelial cells in rats with hepatic fibrosis.72 male rats of clean grade Wistar were randomly divided into 6 groups by random number table method:Normal group, model group, positive control group (Fuzheng huayu capsules), JIANPI RUANGAN DECOCTION of high, medium and low dose group,12 in each group. Rats in normal group were injected with intraperitoneal injection of 30% CCL4 and 70% olive oil mixture,2mL/kg,2 times/ week, and 4 weeks after intraperitoneal injection of normal saline. At the same time, administration from first weeks, the model group and the normal group were given normal saline, the positive control group with Fuzheng Huayu Decoction, Jianpi Ruangan Decoction of high, medium and low dose group were given different doses of Jianpi Ruangan Decoction orally. At the end of the fourth week, using HE staining and Mallory staining of liver tissue fibrosis, light microscope morphological changes of liver fibrosis model group (P< 0.05), indicating that CCL4 induced liver fibrosis rat model is successful; compared with the model group, positive control group, Jianpi Ruangan Decoction high, medium and low dose group of liver fibrosis were reduced (P<0.05).Then, transmission electron microscopy showed that Jianpi Ruangan Decoction high, medium and low dose group and positive control cells fenestra of hepatic sinusoidal endothelial rats, decrease in the number of smaller diameter, reduced somewhat higher than the model group, but the shape does not determine whether there are differences; The content of serum HA detected by ELISA showed that Jianpi Ruangan Decoction high, medium and low dose group can reduce the HA content (P<0.01), and the HA of the high, medium and low dose group decreased with the increase of the dose, showing a certain drug dependence; Real-time PCR and immunohistochemical ABC method to detect the expression of CD31, CD44 and mRNA protein, showed that Jianpi Ruangan Decoction of high, medium and low dose group can increase the positive expression of mRNA and CD44 (P<0.05), CD31 was lowered and the expression of mRNA (P<0.05), and the expression increased with the dose decreased, to a certain extent, drug dependence. All these results indicated that Jianpi Ruangan decoction can improve the effects of liver sinusoidal endothelial cells of rat liver fibrosis defenestrated changes, the mechanism is probably jianpiruangan Decoction and up regulation of mRNA and down-regulation of mRNA CD44 expression of CD31, decrease the content of HA, decrease the rat liver sinusoidal endothelial cell window hole diameter becomes smaller, Decrease in quantity.
