| Objective:To observe the changes of I type,type Ⅲ collagen and fibronectin content in liver tissue and serum of rats with hepatic fibrosis,and to explore the intervention effect of Jianpi Ruangan Decoction on hepatic fibrosis induced by carbon tetrachloride in rats,to investigate the mechanism of liver fibrosis and the therapeutic effect of Jianpi Ruangan Decoction on liver fibrosis.Methodology:1.Animal selection and grouping: After one week of adaptive feeding,the rats were randomly divided into 6 groups,including blank group,model group,Fuzheng Huayu Decoction group,low dose group of Jianpi Ruangan Decoction,middle dose group of Jianpi Ruangan Decoction,high dose group of Jianpi Ruangan Decoction,and each group had 15 rats.2.Model preparation : Rats were intraperitoneally injected with 30%carbon tetrachloride olive oil solution by 0.2ml/100 g,2 times a week,a total of 4weeks.For ordinary feed,free drinking water.3.Prescription intervention: Normal group and model group were perfused with saline administration;Fuzheng Huayu capsule as positive drug on CCl4 induced liver fibrosis in rats by intragastric administration;the Jianpi Ruangan Decoction and divided into high,middle and low dose groups,and they were intragastric administration respectively on CCl4 induced liver fibrosis in rats.1.Index detection: HE staining and collagen fiber staining were used to observe the pathological changes of liver tissue.Immunofluorescence method was used to observe the expression of type I and type Ⅲ collagen and fibronectin in model liver.ELISA was used to calculate the expression of type Ⅰ and type Ⅲcollagen Fibronectin concentration.Results:1.Modeling situation: Compared with the normal group,the serum levels of ALT and AST in model group were increased,the difference possess statistically significant(p<0.05).1.Col I,Col Ⅲ,FN content: ELISA,compared with the normal group,the serum levels of Col Ⅰ,Col Ⅲ,FN in the model group were higher than those in the normal group,and the difference possess statistically significant(p<0.05);Immunofluorescence staining for area observation,compared with the normal group,the expression level of ColⅠ,ColⅢ and FN in the model group was significantly higher than that in the normal group,and the difference was statistically significant(P <0.05);Immunofluorescence staining for gray level observation,compared with the normal group,the expression level of ColⅠ,ColⅢ and FN in the model group was significantly higher than that in the normal group,and the difference possess statistically significant(P <0.05).2.The effect of Jianpi Ruangan Decoction on the contents of Col I,Col Ⅲ,FN: In ELISA,compared with the model group,the serum levels of ColⅠin the Jianpi Ruangan Decoction high dose,middle dose and low dose groups were significantly lower than those in the model group,and the difference was statistically significant(P <0.05);The content of serum ColⅢ in the Jianpi Ruangan Decoction high dose,middle dose and low dose groups were significantly lower,and the difference was statistically significant(P <0.05);The serum levels of FN in the Jianpi Ruangan Decoction high dose,middle dose and low dose groups were significantly lower,and the difference was statistically significant(P <0.05).The liver tissue type I,type Ill collagen and fibronectin were observed by immunofluorescence staining.The expression of the high dose group of Jianpi Ruangan Decoction was lower than that of the middle dose group,and the expression of the low dose group was enhanced,which was more obvious around the central vein.Conclusion:1.A rat model of hepatic fibrosis was successfully replicated by intraperitoneal injection of 30% carbon tetrachloride-olive oil solution twice a week for 4 weeks.2.After the treatment of Jianpi Ruangan Decoction,the levels of type Ⅰ and type Ⅲ collagen and fibronectin in serum and liver of model rats showed that Jianpi Ruangan Decoction could reduce the degree of hepatic fibrosis,and Fibrosis had a good effect. |
