Part One: Effects of different stages aerobic exercise on postoperative liver cancer mice general status and immune functionObjective: By setting different time points of aerobic exercise intervention,four weeks before surgery and seventh days after operation for the movement time to intervene,to explore the different stages of aerobic exercise intervention on general state and immune function effect on mice model difference and related mechanism.Methods: The mice were resection of liver tumor surgery and building the motion model,observation of mice at postoperative spontaneous activity,liver and kidney function,mortality,pulmonary metastase,viscera index,the peripheral blood and spleen lymphocyte CD4 + / CD8 + ratio,detection of spleen lymphocyte proliferation ability,western blot to detection of spleen lymphocytes and explore related molecular mechanism.Results: 1.The total distance for spontaneous activity of normal group is less than normal group,the differences is statistically significant(P < 0.05);Late exercise group and whole exercise group of total distance are greater than control group,except late exercise group,the difference is statistically significant(P < 0.01).2.The ALT and AST between groups of mice are higher than normal group,except control group,there are no statistically significant difference between groups.The TP and A/G ratio between groups of mice are less than normal group.Compared between groups of mice and normal group of blood urea nitrogen,there are no statistically significant difference.Compared to normal mice,the difference in blood uric acid of control group is statistically significant(P < 0.01),and there are no statistically significant difference between groups.Early exercise group,late exercise group and whole exercise group of blood uric acid in mice are lower than control group,differences are statistically significant(P < 0.01).3.The mortality and pulmonary metastase between exercise groups of mice are less than control group.4.Each group of spleen index in mice are greater than normal group,expect late exercise group and whole exercise group,differences are statistically significant(P < 0.01).Each group of thymus index in mice are greater than normal group,there are no statistically significant differences.5.Control group mice peripheral blood and spleen T lymphocytes CD4 + / CD8 + ratio are less than normal mice,the difference is statistically significant(P < 0.05).Late exercise group and whole exercise group compared with normal group,there are no statistically significant differences.Whole exercise group mice peripheral blood and spleen T lymphocytes CD4 + / CD8 + ratio are greater than control group,differences are statistically significant(P < 0.05).The lymphocyte proliferation between groups of mice are higher than normal group,differences are statistically significant(P < 0.05).6.The expression of Beclin1 in each exercise group is higher than control group,and the expression of LC3-? in early exercise group and late exercise group is higher than control group.The expression of Bcl-2,anti-apoptotic proteins,in each exercise group is higher than control group,and the expression of Bcl-x L,anti-apoptotic proteins,is higher than control group.The expression of Caspase 3 and Cleaved Caspase 3 in control group is higher than other groups of mice.Conclusion: Different stages of aerobic exercise intervention can improve the general condition and the immune function of postoperative liver cancer mice in a certain extent,and effects of exercise intervention before surgery combined with postoperative is most significant.Different stages of aerobic exercise intervention can promote the proliferation of spleen lymphocytes in mice,which may be related to inhibit apoptosis and promote autophagy of spleen lymphocytes.Part Two: Effects of aerobic exercise joint sijunzi decoction on postoperative liver cancer mice general status and immune functionObjective: by setting up the postoperative aerobic exercise joint sijunzi decoction intervention program to study the effects and mechanism of aerobic exercise joint sijunzi decoction on the general state and immune function of mice.Methods: the mice were resection of liver tumor surgery,using aerobic exercise and sijunzi decoction combined intervention,observation of mice at postoperative spontaneous activity,liver and kidney function,mortality,pulmonary metastase,viscera index,the peripheral blood and spleen lymphocyte CD4 + / CD8 + ratio,detection of spleen lymphocyte proliferation ability,western blot to detection of spleen lymphocytes and explore related molecular mechanism.Results: 1.In addition to exercise group,the total distance for spontaneous activity between other groups of mice are less than normal mice,differences are statistically significant(P < 0.01);Chinese traditional medicine group,exercise group and exercise combined with Chinese traditional medicine group of total distance are greater than control group,differences are statistically significant(P < 0.01).2.The ALT and AST between groups of mice are higher than normal group,differences are statistically significant(P < 0.01);Chinese traditional medicine group,exercise group and exercise combined with Chinese traditional medicine group of ALT and AST are lower than control group,differences are statistically significant(P < 0.01).Chinese traditional medicine group,exercise group and exercise combined with Chinese traditional medicine group of TP are higher than control group,differences are statistically significant(P < 0.01).The A/G ratio between groups of mice are less than normal group,differences are statistically significant(P < 0.01).Compared to normal mice,the difference in serum urea nitrogen of exercise combined with Chinese traditional medicine group is statistically significant(P < 0.05),and there are no statistically significant difference between groups.3.Chinese traditional medicine group,exercise group and exercise combined with Chinese traditional medicine group of mortality and pulmonary metastase are lower than control group.4.Each group of spleen index of mice are greater than normal group,differences are statistically significant(P < 0.01);Groups of thymus index are greater than normal group,there are no statistically significant differences.5.Sijunzi decoction and exercise intervention can improve peripheral blood and spleen lymphocytes in mice CD4 + / CD8 + ratio and promote spleen lymphocyte proliferation.Sijunzi decoction combined with aerobic exercise intervention group has the optimal effect.6.The expression of Beclin1 and LC3-? in exercise group and exercise combined with Chinese traditional medicine group are higher than control group,the expression of Beclin1 and LC3-? in Chinese traditional medicine group are not obviously rise.The expression of Bcl-2 and Bcl-x L,anti-apoptotic proteins,in Chinese traditional medicine group are higher than control group,and the expression of Caspase 3 and Cleaved Caspase 3 are lower than control group.The expression of Bcl-2 in exercise group is higher than control group,and the expression of Caspase 3 and Cleaved Caspase 3 are lower than control group.The expression of Bcl-2 in exercise combined with Chinese traditional medicine group is higher than control group,but the expression of Bcl-x L,Caspase 3 and Cleaved Caspase 3 are no obvious difference in control group.Conclusion: Sijunzi decoction intervention and postoperative aerobic exercise intervention can improve the general condition and immune function of postoperative liver cancer mice in a certain extent,and effects of aerobic exercise combined with sijunzi decoction intervention group is the best.Sijunzi decoction intervention can promote the proliferation of mouse spleen lymphocytes,which may be related to inhibit apoptosis;sijunzi decoction combined with aerobic exercise intervention can promote lymphocyte proliferation,which may be related to promote autophagy.And the specific mechanism needs to further research.
