Part One: Effects of aerobic exercise of different intensities on hepatocarcinoma formation in DEN induced miceObjective: Taking aerobic exercise as an intervention,setting 12 m / min and 18 m / min speed on DEN induced mice,observing the general state of mice,detecting related indexes of liver and kidney function,the rate of liver cancer and the death rate of mice,and identifying the mortality of mice induced by DEN and the rate of HCC tumor formation.It provides experimental evidence for inhibiting the formation of liver cancer by aerobic exercise.Methods: By intragastric administration of DEN induced mouse model of liver cancer,and exercise intervention of different intensity on mice,observing the differences of liver function,renal function,mortality,the incidence of liver cancer of mice.Results: 1.Compared with the blank group,the ALT and AST in DEN group were higher,and the difference was statistically significant(ALT:40.40±5.63 VS 110.70±7.08;AST:140.90±21.89 VS 677.00±77.47;P<0.01).Compared with the DEN group,the ALT and AST decreased significantly in the medium speed group and in the high speed group,and the difference was statistically significant(ALT:110.70±7.08 VS 64.11±7.27;110.70±7.08 VS 61.13±6.62;AST: 677.00±77.47 VS 208.50±43.45;677.00±77.47 VS 198.9±45.69;P<0.01).2.Compared with the blank group,the BUN and UA in the DEN group were significantly higher,and the difference was statistically significant(BUN: 3.055±0.798 VS 9.875±1.089;UA: 114.40±29.54 VS 317.30±43.26;P<0.01).Compared with the DEN group,the UA in the medium speed group and the high speed group decreased,and the difference was statistically significant(317.30±43.26 VS 192.0±38.59;317.30±43.26 VS 199.9±40.39;P<0.01).Compared with the DEN group,there was no significant difference between the medium speed group and the high-speed group.3.there was no significant difference in mortality between the blank group,DEN group,medium speed group and high speed group.4.Compared with the blank group,the incidence of liver cancer in the DEN group was significantly higher,and the difference was statistically significant(0% VS 94.73%;P<0.05).Compared with the DEN group,the incidence of liver cancer in the middle speed group and the high speed group was significantly lower,and the difference was statistically significant(94.73% VS 66.67%;94.73% VS 56.25%;P<0.05).Conclusion: Aerobic exercise can significantly improve the general condition of DEN induced liver cancer mice.Aerobic exercise can reduce the mortality and the incidence of liver cancer induced by DEN.Part Two: Effects of aerobic exercise on iron metabolism in mice induced by DENObjective: By observing the difference of iron metabolism related indexes in each group,to demonstrate the correlation among the three groups of aerobic exercise,iron metabolism and the formation of liver cancer and whether aerobic exercise could inhibit the occurrence of liver cancer in DEN induced model mice by affecting iron metabolism.Methods: On the basis of the first part of the experiment,iron,Ferritin and Hepcidin were detected in the serum,muscle tissue and liver tissues of each group,and the differences in the expression of TfR1,TfR2 and FPN1 on the membrane of the myocytes were observed to confirm whether aerobic exercise could inhibit the occurrence of liver cancer in the DEN model mice by promoting the iron metabolism.Results: 1.Compared with the blank group,the serum Fe level of DEN group increased significantly(23.71 ± 9.895 VS 54.86 ± 5.383;P<0.01).Compared with DEN group,the serum Fe level in medium speed group decreased significantly,and the difference was statistically significant(54.86 ± 5.383 VS 29.31 ± 9.997).Compared with the blank group,the serum Ferritin level of DEN group increased significantly,and the difference was statistically significant(1944 ± 439.6 VS 3799 ± 685.5;P<0.01).Compared with DEN group,the serum Ferritin level in medium speed group decreased significantly,and the difference was statistically significant(3799 ± 685.5 VS 2318 ± 555.4;P<0.01).Compared with the blank group,the serum Hepcidin content in the DEN group was significantly higher,and the difference was statistically significant(239.2 ± 16.90 VS 318.1 ± 28.80;P<0.01).Compared with the DEN group,the serum Hepcidin content in the medium speed group was significantly higher,and the difference was statistically significant(318.1 ± 28.80 VS 512.7 ± 26.35;P<0.01).2.Compared with the blank group,the iron content in the muscle tissue of the DEN group was significantly lower,and the difference was statistically significant(3.117 ± 0.325 VS 2.876 ± 0.232,P<0.05),and the iron content in the muscle tissue of the DEN group was significantly higher than that in the medium speed group,and the difference was statistically significant(2.876 ± 0.232 VS 4.199 ± 0.313;P<0.01).Compared with the blank group,the content of Ferritin in the muscle tissue of the DEN group increased significantly(13.32 ± 0.909 VS 17.34 ± 1.416,P<0.01).Compared with the DEN group,the Ferritin content of the muscle tissue in the medium speed group decreased significantly(17.34 ± 1.416 VS 11.01 ± 1.330;P<0.01).Compared with the blank group,there was no significant change in the content of Hepcidin in the muscle tissue of the DEN group.Compared with the DEN group,the Hepcidin content of the muscle tissue in the medium speed group was significantly higher,and the difference was statistically significant(48.83 ± 3.409 VS 81.67 ± 7.090;P<0.01).3.Compared with the blank group,the Fe content in liver tissue of DEN group increased significantly,and the difference was statistically significant(5.007 ± 0.738 VS 8.118 ± 0.865;P<0.01).Compared with DEN group,Fe in liver tissue of medium speed group decreased significantly,with a statistically significant difference(8.118 ± 0.865 VS 4.997 ± 0.589;P<0.01).Compared with the blank group,the liver tissue Ferritin content in the DEN group was significantly increased(28.79 ± 4.093 VS 41.89 ± 3.069,P<0.01),and the Ferritin content of the liver tissue in the medium speed group was significantly lower than that in the DEN group,and the difference was statistically significant(41.89 ± 3.069 VS 26.71 ± 4.567;P<0.01).Compared with the blank group,there was no significant difference in the content of Hepcidin in the liver tissue of the DEN group.Compared with the DEN group,the Hepcidin content in the liver tissue of the mice was significantly higher in the medium speed group(4.492 ± 0.776 VS 10.290 ± 0.867;P<0.01).Conclusion: Aerobic exercise increases the intake of iron of muscle tissue during exercise,and the level of Hepcidin in the body,and decrease the level of serum Fe and Ferritin in the body,which may be related to the occurrence of liver cancer in mice inhibited by aerobic exercise.
