Objective To construct an oxygen-induced retinopathy mouse(OIR)model to observe the inhibitory effect of Panax notoginseng saponins on oxygen-induced neovascularization of retinopathy,and investigate the effect and possible mechanism of Xuesaitong on neovascularization in retinopathy of premature infants.Methodology Use new C57 BL / 6J mice were divided into 5 groups(n = 15).Group A was the normal control group,that is,the mice were kept in the natural environment,group B was oxygen-induced retinopathy model group(OIR group),group C was OIR + Panax notoginseng low dose intraperitoneal injection group,group D was OIR+ Total saponins in high dose intraperitoneal injection group,E group was OIR +Panax notoginseng saponins glass chamber treatment group.Retinal tissue was collected in P17.In every group,P17 retinal tissue,Fluorescein isothiocyanate dextran(Fuorescein isothiocyanate,FITC-dextran),HE staining;reverse transcriptase polymerase chain reaction(RT-PCR)detection of vascular endothelial growth factor(Vascular endothelial growth factor A,VEGFA),hypoxia inducible factor-1?(Hypoxia-Inducible alpha Factor1,alpha HIF-1?)expression;Western blot method to detect VEGFA,HIF-1?,protein kinase C(protein kinase C,PKC)expression.Results Oxygen-induced model of retinopathy in mice was successfully constructed.The growth and development of mice in OIR model group were significantly slower than those in normal air group.The mice in the treatment group were well developed.The results of retinal deposition and HE staining showed that the OIR model group compared with the normal air group retinal blood vessels,the retinal blood vessels were thickened,bent,with large fluorescence leakage and neovascularization;treatment group of vitreous cavity injection group on retinal neovascularization inhibition More pronounced,retinal vascular thickening,bending improved significantly.RT-PCR was used to detect the expression of VEGFA and HIF-1? m RNA in the retina.The expression of VEGFA and HIF-1? m RNA in the intravitreal injection group was significantly decreased in the OIR model group.The expression of VEGFA,HIF-1? and PKC in OIR model group was significantly higher than that in control group.The expression of VEGFA,HIF-1? and PKC protein in the intravitreal injection group were significantly decreased.Conclusion Panax notoginseng saponins can improve the oxygen-induced neovascularization of retinopathy and inhibit the expression of VEGFA,HIF-1? and PKC.
