| Objective:Observing how PuQin enema method works in curingulcerative colitis(UC),knowing the enema's influence to the amount of TNF-?,IL-2,IL-10,and probing for the concrete mechanism.Method:Randomly divided 48 healthy SD ratsinto TCM(PuQin enema)original dose group,TCM low dose group,mesalazine group and model group,there're ten rats in each group above and there,re 8 rats in sedentary control group.Establish the UC rats model by using trinitrobenzenesulfonic acid(TNBS),then clyster normal amount of TCM to original dose group,half normal amount of TCM to low dose group,mesalazinesuspension to mesalazine group and 0.9%NS to model group.Sedentary control group are normally fed.Check the weight,feces,feeding and drinking of rats every day,after 2 weeks execute the all rats,keep the serum and colon tissue samples and measure the amount of IL-2,IL-10,TNF-? in serum and tissues by ELISA.At the same time,observe the colonic histological changes.Result:1.The rat's general status and disease activity index(DAI)Compared to sedentary control group,the DAI of model groups obviously get higher and after curing the DAI of three healing groups descend and are all significantly different from the model group(P<0.01).Symptoms improved more rapidly in the original dose group,and is aslo significantly different from the low dose group and mesalazine group(P<0.01).2.Observation of pathological histology colonunder light microscopeWe can see mucosal hyperemia edema and ulcers in model group and with inflammatory cells invasion.It's histological score is significantly higher than the other groups which is significantly different from other groups(P<0.01).After using the medicine the inflammatory of the rats have been improved and the histological scores significantly descend,but there are noobvious difference among the three treatmentgroup(P>0.05).3.the amount of IL-2,IL-10,TNF-a in serum and tissuesTNF-? in serum and tissues in model group get higher than sedentary control group and the amount of IL-2,IL-10 descend which is significantly different from sedentary control group(P<0.01).After treatment the TNF-a in serum and tissues in three treatment groups decline significantly and the amount of IL-2,IL-10 have got higher.Three treatment groups are all significantly different from the model groups(P<0.01).Within three treatment groups,TNF-a in serum and tissues in original dose group get obviously higher than mesalazine group,and the amount of IL-2,IL-10 declined obviously compared to the mesalazine group.There is significant difference between two groups(P<0.01).And the amount of IL-2 in original dose group get obviously higher than low dose group(P<0.01).The others have no statistical significance(P>0.05).Conclusion:PuQin enema can adjust the balance of the cytokine network and improve the body immunity,effectively alleviate the intestinal inflammation of UC rats model and accelerate the process of ulcer healing.The effect is related to the dose of TCM and the main advantage is that it can improve the level of IL-2,rapidly improve the UC rats clinical symptom. |
