Obiective:To investigate the protective mechanism of the extract of Bidens pilosa L.(Bp)on ocular surface and lacrimal gland of androgen deficiency dry eye.Methods:Twenty-four female Wistar rats were randomly divided into four groups:Group Control?Group Saline?Group Model and Group Bp,each group 6 rats.Group Control did not do any treatment.Group Saline gavage physiological saline(0.5ml/0.1kg),once per day,For 4 weeks.Group Model gavage finasteride(1.16mg/kg/day),once per day,For 4 weeks.Group Bp gavage finasteride and BP(0.5ml/0.1kg),once per day,For 4 weeks.The experimental data collection and specimen collection time:each group was measured with phenol red-impregnated cotton threads?tear film breakup time and corneal fluorescein staining at 0 day?7day?14day?21day and 28day.Animals were sacrificed at 28 days,gene microarray analysis for inflammatory cytokines of lacrimal gland and cornea,according to the results of gene microarray analysis,tests the Western blot of lacrimal gland.Lacrimal gland and corneal tissue were evaluated with light microscopy.Results:?Group Fin rats had significantly lower aqueous tear quantity?BUT and higher fluorescein staining scores than the group Con rats,the lacrimal gland had significant inflammatory cell infiltration.Compared with the Fin group,the aqueous tear quantity?BUT significantly improved and lower fluorescein staining scores with Bp treatment in the group Bp rats,and the structures of the lacrimal gland were well maintained without significant lymphocyte infiltration.?Using cytokine antibody arry technology to detect the results show that,expression levels of 22 cytokines in the lacrimal gland were changed significantly in group Fin compared to group Con.Most of these cytokines were considered to be inflammatory factors and chemokines.The expression levels of 15 cytokines(IL-1?,IL-6,IFN-?,Fas L,GM-CSF,TNF-?,IL-4,IL-10, CNTF,B7-2/Cd86,Leptin,MMP-8,TIMP-1,L-selection and ICAM-1)were significantly increased.Among the highly expressed cytokines,IL-1?,IL-6,GM-CSF and TNF-? are proinflammatory cytokines,and IL-4 and IL-10 are anti-inflammatory genes.Meanwhile,the expression of those cytokines decreased significantly in group Bp compared to group Fin.When compared with group Con,the expression of almost all cytokines in group Bp showed no significant differences;only several cytokines were moderately increased.The expression levels of 6 cytokines in the cornea were significantly elevated in group Fin compared to group Con,including Fsa L,IL-13,PDGF-AA,B7-2/Cd86,TIMP-1 and Prolactin R.The expression of almost all cytokines in group Bp showed no significant differences when compared to group Con.?Western blot analysis of high expression of cytokines in lacrimal gland,including(B7-2/Cd86,MMP-8,TIMP-1,IL-1?,IL-4,IL-6,IL-10,FasL and TNF-?,the expression levels of MMP-8,IL-1?,IL-4,IL-6,IL-10,Fas L and TNF-? were significantly increased in group Fin compared to group Con(P<0.05 or P<0.01).B7-2/Cd86 and TIMP-1 were also increased with no statistical significance.Compared with group Fin,IL-1?,Fas L,and TNF-? in group Bp were significantly decreased(P<0.05).Other cytokines also showed this decreased tendency but without statistical significance.Conclusion:Finasteride administration effectively induced dry eye in rats by 14 days after administration.Through the treatment of BP can effectively relieve androgen deficiency dry eye symptoms,improve tear secretion,stable tear film,inhibition of lacrimal gland inflammation.Biden grass has a good application prospect in the treatment of androgen deficiency dry eye.
