Study On Reparation Process And Quality Standard Of Lurongduanxu Cataplasm

Lurongduanxu Cataplasm is a new traditional Chinese medicine,which consists of four kinds of herbs that are antler velvet,radix dipsaci,drynaria fortune and panax notoginseng.It has the effect on promoting the rate of bone growth,improving the fracture healing and reducing the fracture complication.It is mainly used for the treatment of broken bones.This subject is based on Chinese medicine theory and has completed a major new drug research of Lurongduanxu Cataplasm,including the preparation process,quality standard,stability and metabolites research.1.Study on preparation processThe extraction process of antler velvet: we tested different concentrations of ammonia-ammonium acetate in buffer,the liquid–solid ratios and ultrasonic power to influence protein content of antler velvet extraction with the orthogonal test,and obtained the proper process conditions.In the study on quality standard of antler protein,we authenticated the protein by the application of TLC and determined the content of IGF-1 by radioimmunoassay.The preparation process of Lurongduanxu Cataplasm: the kinds of matrixes and range of dosages were determined on the base of preliminary experiments,with the adhesive force,peeling strength and comprehensive scores of sensory evaluation as the evaluation indexes,through the experiment of the orthogonal design.2.Study on quality standard and stabilityThe asperosaponin ? and naringin were authenticated by TLC.The plaster ointment,shape forming,adhesive and microbial limit were researched.The content of asperosaponin ? and naringin were determined by HPLC.According to the relevant provisions in the quality standard,the results of testing indicated the stability of the Lurongduanxu Cataplasm in the storage period.3.Study on the metabolites of Lurongduanxu CataplasmThe metabolites of Lurongduanxu Cataplasm were researched by UPLC/QTOF-MS and UNIFI platform.Then eight metabolites of naringin were identified from mouse blood after sticking Lurongduanxu Cataplasm.4.ConclusionLurongduanxu Cataplasm,which has been developed,is stable,adhesion to skin is good,less amount of remaining on the polyethylene membrane.The simple production process is low cost and easy for industrialized production.Through the extraction process,preparation process,quality standard,stability,the metabolites researching and so on,the study lays a good foundation for further clinical application of Lurongduanxu Cataplasm.
