Observation Of Morphology And Organization Structure As Well As Study On Distribution Law Of Several Saponins Of Medicinal Organs Of Pulsatilla Chinensis (Bunge) Regel

1 Objective(1)On the basis of Baitouweng materia medica textual research,provided the basis for the reform of geoherbs and the resource utilization of Pulsatilla chinensis;(2)The observation of genuine origin of Baitouweng of Chuzhou city in Anhui province to explore the biological characteristics and growth habit of the Pulsatilla chinensis and provide reference for the identification and quality evaluation of Radix pulsatilla;(3)Be from the medicinal properties and morphological identification of Baitouweng found in character identification;(4)Application of microscopic identification technology to study on Pulsatilla medicinal parts of anatomical and histological structure and Pulsatilla medicinal organ development,explore,Provided the basis for the identification of Pulsatilla chinensis;(5)On the basis of Baitouweng organizational structure,choosing the content of saponins in different parts of Baitouweng officinal organs of different diameters,Explore the accumulation of saponins in Pulsatilla medicinal organs of different diameter and different parts and different organizational structures,to provide reference for harvesting and processing and quality evaluation of Pulsatilla medicinal materials.2 Method(1)Summarize the records of the Baitouweng and its producing areas in the herbal books;(2)By using the fixed point and periodic observation methods of wild Pulsatilla chinensis in Chuzhou province,development and biological characteristics were studied,the growth habit of wild Pulsatilla chinensis proved,to provided reference for the cultivation and quality evaluation;(3)Different parts of Pulsatilla medicinal materials: Baitouweng petiole residues and buds,joints and the joints on the lower part of festival festival,to provide a basis for the identification of medicinal materials of Pulsatilla chinensis;(4)The microscopic identification of primary structure and secondary structure of Baitouweng development.Medicinal organs ofdifferent diameter,different parts of the anatomy of the structure,provide a basis for the identification of clear composition Pulsatilla medicinal parts;(5)In the early stage based morphological and microscopic identification,determination of different diameter,different parts and different organizational structure of the medicinal organs Pulsatilla Saponin B4 and pulchinenoside A3 and pulchinenoside D.Three saponins by HPLC-ELSD method,in order to reveal the distribution of the three kinds of saponins in Radix Pulsatillae accumulation.3 Results(1)On the history of textual research shows that since the Song Dynasty,Baitouweng origined Ranunculaceae Baitouweng genuine Pulsatilla chinensis(Bunge)Regel,consistent with this.Since the beginning of the Ming Dynasty geoherbalism Chuzhou Baitouweng attention,after the Republic of China in Chuzhou province;Tang and Ming Dynasties based in raw materia medica,Baitouweng also may include p.cernua,p.dahurica,p.ambigua and other Chinese pulsatilla.In this paper,suggestions on the in-depth study of Chuzhou Baitouweng,the development of high-quality Baitouweng resources;suggestions p.cernua,p.dahurica,p.ambigua should place the drug habit;(2)The germination of Chuzhou wild Pulsatilla in mid February,late February to enter the flowering,fruit period for 4-5 months,6-7 months to maintain growth stage,September start nutrition growth,nutrient accumulation,11-12 months for down seedlings.(3)The existence of underground organ of Pulsatilla joints,joints and underground organ of the fine is obvious.At the same time,it has the Pharmacopoeia" to suggest the modified Pulsatilla medicinal parts.The secondary structure of Pulsatilla is not developed,no cork tissue,composed of epidermis,cortex,phloem in the exercise of protection,growth in the root of the secondary stage,endothelial layer still exists,with the tangential splitting capability of the cell,to protect the function of the organization.(5)The content of saponins showed that little change in content in different diameter of Baitouweng in the underground organs of pulchinenoside B4.Pulsatilla Saponin A3 and pulchinenoside D content in Radix Pulsatilla larger diameter decreased.Determination of Pulsatilla medicinal organ in different parts of the saponins content of pulchinenoside B4 found that the highest content in the rhizome part of root root was the lowest;secondly,pulchinenoside A3 was the highest in the rhizome and root content,the lowest content in roots;content of Pulsatilla Saponin D highest in taproot,lateral root of the rhizome was the lowest.Determination of the content of saponins in different structure of medicinal organs in Baitouweng,pulchinenoside B4,pulchinenoside A3 and pulchinenoside D was the highest content of cortex,xylem was the lowest.4 ConclusionHerbalogical study showed with the original Baitouweng medicinal Pharmacopoeia based Pulsatilla chinensis(Bge.)Regel.The morphology of the original plants of wild Pulsatilla chinensis showed that there were joints in the medicinal parts,and the microscopic identification showed that the above part of the joint is a rhizome,the root below the joint point is the root.The origin of Bozhou medicine market investigation and survey found that the joint as the main part of Pulsatilla medicinal materials.Change Baitouweng pulchinenoside B4 content herbs of different diameter little,3 kinds of saponin content of Pulsatilla medicinal parts of different parts and different organizational structure has difference.The highest content of saponin in the root stem of bald head is in line with the current harvesting and processing methods.
