Effects Of Yishenhuoxue Recipe On Sperm Apoptosis And DNA Integrity Of Patients With Kidney Deficiency And Blood Stasis Type Of Infertility

ObjectiveTo observe the effects of Yishenhuoxue Recipe on sperm quality(semen volume;sperm concentration;sperm motility;forward motile sperm percentage;normal sperm percentage),sperm apoptosis and DNA fragmentation index(DFI)of the patients with kidney deficiency and blood stasis type of infertility before and after treatment.To make an objective evaluation of the clinical effects about Yishenhuoxue Recipe.MethodsCollecting cases from reproductive center in the first affiliated hospital of guangzhou university of TCM from April,2016 to January,2017.50 patients with kidney deficiency and blood stasis type of infertility were randomized into two groups.Among the 50cases,8 cases were dropped because of not according to the prescribed medication and not completing the treatment cycle.42 patients were included in the analysis:22 cases in Chinese traditional medicine group(Yishenhuoxue Decoction group),20 cases in control group(tamoxifen and Vitamin E tablets group).The day patients got initial diagnosis were considered as the start date of treatment and observation,continuous observation for 90 days.Main parameters of semen quality,PAS and DFI before and after treatment of the two group were recorded after 12 weeks.The 4th week after the treatment,the pregnancy situation of the patient's spouse was followed up.We used SPSS19.0 to analyze datas,just P<0.05 considered as significant.ResultsBefore the treatment,the age,infertile time,main parameters of semen quality(semen volume;liquefaction time;sperm concentration;sperm motility;forward motile sperm percentage;normal sperm percentage),sperm apoptosis and DNA fragmentation index(DFI)have no obvious difference(all the P>0.05).After the treatment,both of the two groups show significance that they could increase patients semen volume;reduce the patients' semen liquefaction time;improve patients' sperm concentration,sperm vitality,forward motile sperm percentage and normal sperm percentage;and decrease sperm apoptosis rate and DFI(P<0.05).On the sperm vitality,forward motile sperm percentage,normal sperm percentage,and sperm apoptosis,Chinese medicine group is superior to the control group.By t test,it shows statistically significant difference(P<0.05).Among 22 cases in the Chinese traditional medicine treatment group,the cured case,significantly effective case,effective case,ineffective case is respectively 2,8,10,2,the total effective rate is 90.91%;among 20 in the control group,it is respectively 0,5,7,8.The total effective rate is 60%.By rank sum test,the Chinese traditional medicine treatment group curative effect is better than that of control group,the difference was statistically significant(P<0.05).By X2 analysis,the Chinese traditional medicine treatment group is superior to the control group,the difference was statistically significant(P<0.05).Conclusion1.Yishenhuoxue Decoction can effectively improve the pregnancy rate of Patient' s spouse in treating of the patients with kidney deficiency and blood stasis type of infertility.And the significance is remarkable,compared to control group.2.Yishenhuoxue Decoction is superior to the control group on effectively improving patients' sperm vitality;forwarding motile sperm percentage and normal sperm percentage;decreasing sperm apoptosis rate.3.Kidney deficiency and blood stasis is an important reason for infertility patients.The method of nourishing kidney and activating blood is an important method in treating infertile patients.We can choose Yishenhuoxue recipe as a traditional Chinese medicine treatment method to decrease sperm apoptosis and DFI.
