Research backgroundIn recent years,the incidence of infertility is on the rise,in which the proportion of patients with dysspermia is gradually increasing;dysspermia is relatively common in clinical practice,and its known causes are complex and diverse,but its pathogenesis is not very clear,and the treatment has not reached an agreement.Modern medicine has no targeted treatment drugs,and traditional Chinese medicine has achieved good results in the treatment of this disease.Wang Yiguang,a tutor,thinks that the main pathogenesis of dysspermia is kidney deficiency,which includes deficiency of kidney qi,deficiency of kidney yin and deficiency of kidney yang.The kidney qi promotes the normal operation of Tiangui,the kidney Yin nourishes the kidney essence,the kidney yang is warm and promotes the formation of sperm.The tutor thinks that the treatment of dysspermia can be based on the macroscopical syndrome differentiation of traditional Chinese medicine and the etiology of Western medicine.In the treatment of traditional Chinese medicine,the syndrome differentiation of dysspermia is often the pathological factors of dampness heat,dampness heat accumulation and blood stasis for a long time,which eventually lead to dysspermia.Therefore,the tutor thinks that the dysspermia is based on the deficiency of kidney,with damp heat as the standard.In the treatment,we should keep the pathogenesis in mind,invigorate the kidney,clear the heat and remove the dampness.According to the situation,we should promote blood circulation.In the clinic,we often choose the prescription of invigorating the kidney and removing the dampness to treat the dysspermia,and get a better effect.In order to further study the mechanism of Bushen Lishi formula,and provide experimental basis for clinical practice,we selected this formula for experimental study,in order to clarify the way to improve the deformity of spermatozoa,and provide basis for later treatment.Research objectiveIn this experiment,Tripterygium wilfordii polyglycoside was used to establish the model of spermatogenesis disorder in rats.By observing the general situation,semen quality,sperm morphology,testicular pathology,the content of spermatoprotein and its mRNA expression of rats with spermatogenesis disorder,to explore the influence of Bushen Lishui formula on the above indexes,and to explore its mechanism preliminarily,so as to provide experimental basis for clinical practice.Research method60 mature male SD rats of SPF level were randomly divided into 2 groups,50 in the experimental group and 10 in the blank control group.The experimental group was randomly divided into model group,positive control group,low dose group,medium dose group and high dose group,with 10 rats in each group.The blank control group was given 18ML/kg normal saline for 8 weeks;the other groups were given 30mg/kg·D Tripterygium polyglycosides for 8 weeks;the positive control group was given vitamin E at the fifth week 25 mg/kg body weight was given to the stomach,and the high,medium and low dose groups were given Bushen liwet formula(raw Rehmannia 15g,Cornus 12g,yam 12g,Poria 15g,Danpi 12g,Alisma 9g,Phellodendron 9g,Astragalus 20g,stir fried Atractylodes 15g,dodder 9g,Ligustrum 12g,shayuanzi 12g,Epimedium 9g,plantain 12g,raspberry 12g,Kui 9g,honeysuckle 9g,Hedyotis 12g)concentrate 9,18 27g crude drug/kg body weight,once a day for four weeks.The rats in each group were killed 24 hours after the last administration,and the semen routine,sperm morphology,testicular pathology,sperm nuclear protein content and mRNA expression were detected.Research results1.There was no significant difference in the body weight of rats in each group(P>0.05);the body weight ratio of testis in high dose group was significantly higher than that in low dose group,positive control group,model group and blank group(P<0.05).2.The sperm activity of high dose group was significantly higher than that of model group and low dose group,and that of blank group was significantly higher than that of low dose group and model group(P<0.05);the sperm density of high dose group was significantly higher than that of low dose group,positive control group and model group,and that of blank group was significantly higher than that of low dose group,positive control group and model group(P<0.05)The rate of abnormal sperm in type B group was significantly higher than that in other groups(P<0.05).3.In addition to the blank group,all the other groups had varying degrees of changes in seminiferous tubules,stromal cells and Sertoli cells,and the number and number of spermatogenic cells decreased.According to the degree of injury,the model group>low dose group>middle dose group>positive control group>high dose group>blank group.4.In the model group and the low-dose group,the relative expression of protamine P1 was lower than that in the high-dose group;in all groups except the high-dose group,it was lower than that in the blank group;in the low-dose group and the positive control group,the relative expression of protamine P2 was significantly lower than that in the blank group(P<0.05).5.The expression of P1 mRNA in high dose group was significantly higher than that in control group,low dose group,middle dose group and positive control group;the expression of P2 mRNA in control group was lower than that in blank group;the difference was statistically significant(P<0.05).Conclusion(1)Bushen Lishui formula can improve the structure of seminiferous tubules,protect Leydig cells and Sertoli cells,and reduce the apoptosis of spermatogenic cells in SD rats.(2)Bushen Lishui formula can improve the sperm activity,density and the proportion of abnormal sperm in SD rats.(3)Bushen Lishui recipe can increase the expression of P1 and its mRNA in spermatozoa of SD rats,and reduce the proportion of abnormal spermatozoa by this way.
