Objective: Oral squamous cell carcinoma(OSCC)is one of the ten most common cancers in the world.It not only endangers patients' lives,but also effects cosmetic appearances and quality of lives.Despite improvements in therapeutic approaches such as operation with or without radiochemotherapy,the 5-year overall survival rate of patients with late-stage tumors are only 40% ~50%.However,for patients with early-stage tumors,it rise to 85.4%.Accordingly,early detection of OSCC is important to improve patients' outcomes and it is confirmed that combined detection of tumor markers(TM)are essential for early diagnostic and monitoring of malignant tumors.The objective of this study is to investigate the expression of tumor suppressor gene PTEN and cytokeratin 19(CK19)among Uyghur OSCC patients,determine their clinical significance and role in the development of OSCC.Methods: Pathologically confirmed paraffin embedded tissues from 102 cases of OSCC and 99 cases of oral benign tumor were obtained.Immunohistochemistry EnVision method was used to detect the expression level of PTEN and CK19 proteins,and SPSS 22.0 package was used for statistical analysis to correlate protein expression level with clinical and pathological findings.Results: Decrease or deletion of PTEN expression in OSCC was found(P<0.001),the positive expression rate in OSCC and benign control group were 37.26%(38/102)and 98.99%(98/99)respectively,but the correlation with tumor pathology and clinical stage was not significant.However,the positivity rate of CK19 in OSCC is 43.14%(44/102),and expression was significantly correlated with differentiation,malignancy degree and clinical stage of tumors(P<0.005).The poorer the pathological differentiation grade,the higher the level of CK19 expression.The positive rate of CK19 in control group was not identical in different type of tissues.Conclusion: PTEN and CK19 expression are associated with the occurrence,development and transfer of OSCC,and the two proteins together could be an indicator of prognosis.PTEN and CK19 could be used as complementary tumor markers of OSCC and combined detection of them has certain value in clinical diagnosis and treatment of OSCC.
