Vitamin B2,also known as riboflavin,forms flavin mononucleotide?FMN?and flavin adenine dinucleotide?FAD?in the body,acting as coenzyme.The two coenzymes play an important role in energy metabolism,especially in the metabolism of fat,carbohydrate and protein.Vitamin B6 including pyridoxine,pyridoxal and pyridoxamine,plays an important role in physiological activity metabolic reactions in human and animal body.In addition,vitamin B6 is also involved in the metabolism of fat,reducing the cholesterol in the blood.Vitamin B2 and cooperated with vitamin B6 participates in some metabolic process in the body,such as amino acid metabolism.However,vitamin B2 and vitamin B6 are easily destroyed by light and alkaline enviroment,causing the damage of their chemical structure and reduction of bioactivity which is unfavorable for their application.Soybean meal?SBM?is an ideal food material rich of protein.In this study,SBM through superfine pulverizing firstly and ball mill grinding secondly turn into the nSBM,that readily aggregates into larger particles.It provides a new possibility of the use of vitamin B2 and vitamin B6 in the solution.In this study,our work included constructing the delivery system and the characterization of the delivery system.Then,we used CCK-8 to detect the cytotoxicity of nSBM on Caco-2 cells and the bioavailability of vitamin B2 and vitamin B6 base Caco-2 monolayer.First work of this study is the quantification of vitamin B2 and vitamin B6 in a mixed solution at a high concentration by fluorescence spectrophotometry.Firstly,the determine wavelength of vitamin B2 and vitamin B6 was Ex/Em=480/530 nm and Ex/Em=300/400 nm respectively.A mathematical model was established to calculate the actual concentration of vitamin B6 based on the apparent concentrations of vitamin B6 and vitamin B2,which were quantified directly when vitamin B2 and vitamin B6 were mixed together and R2 is 0.999.By ANOVA,different particle sizes of SBM had significant impact on the loading efficiency of vitamin B6,but no significant influence on vitamin B2.Loading efficiency of vitamin B2 related to the amount of nSBM and vitamin B2,regardless of the amount of vitamin B6.To the contrary,not only the amount of nSBM and vitamin B6 but also the vitamin B2 influenced the loading efficiency of vitamin B6.The release of vitamin B2 in delivery system within 5h were 86.153%,89.585% and 97.761% corresponding the particle size of nSBM 200 nm,500nm,800 nm.However,as for vitamin B6,the release in delivery system within 5h according to the particle size of nSBM were 93.691%,91.741% and 75.941%,respectively.Comparing the IR spectra of vitamin B2 vitamin B6,and nSBM with delivery system,a new characteristic peak?3431.55cm-1/3420.38cm-1?was found,which meant that the hydrogen bond was the major binding force between vitamin B2 or vitamin B6 with nSBM.SEM results show that the nSBM particle turned to spheroidal particle and the particle size was 2 ?m.In CCK-8 detection,cell viability value over 100%,suggesting that nSBM had no effect on Caco-2 cells.The transport of vitamin B2 and vitamin B6 was studied by Caco-2 monolayer cell model.There was no significant difference between the delivery system and free vitamin B2 and vitamin B6.Considered that delivery system could stay longer in the intestinal tract,there were higher bioavailabilities of vitamin B2 and vitamin B6 were higher in delivery system.This study used nSBM to delivery vitamin B2 and vitamin B6 which could enhance the bioavailability of vitamin B2 and vitamin B6.The application of food borne materials is the main innovation of this study.Smaller nSBM particle should be produced with expect to increase the loading efficiency of vitamin B2 and vitamin B6,enhance the stability of the delivery system need to be invested in future work.
