OBJECTIVE MicroRNA is a single-stranded,non-coding small molecule RNA(about 22 nucleotides of non-coding small molecule RNA)that is about21 nt long.Mi RNAs are highly conserved during evolution by specifically binding to the 3 & apos;UTR region of the target gene sequence,inducing target m RNA degradation or repression,miRNAs are expected to modulate more than 60% protein-encoding genes and participate in almost all known cells process.Mi RNAs play an important regulatory role in cell proliferation,differentiation,metabolism,apoptosis and development.Its participation in disease occurrence,development and outcome process,and through the regulation of oncogene or tumor suppressor gene expression to affect the occurrence and development of the tumor.In recent years,serum tumor markers because of its relatively simple,less trauma to the patient,early warning of the diagnosis of malignant tumors more and more favored by the majority of medical researchers.Circulating miRNAs are very stable in plasma and serum,and are not cleaved by RNA cleavage after binding to specific proteins in serum;the stability of miRNAs in plasma and serummakes their actual values The patient's test value is very consistent.Because of this,miRNAs as a tumor marker have great potential.In this study,miRNA-210 was detected in the serum of esophageal cancer patients and normal subjects.The differences of miRNA-210 expression between the observation group and the control group were compared with those of different tissues,staging and grade esophageal cancer.(Mi RNA-210)as a tumor marker to guide the early diagnosis of esophageal cancer,as well as the diagnosis of esophageal cancer type,staging,grading.METHODS Through previous research,serum samples of esophageal cancer and normal human serum miRNA-210 were selected as follows: The first step was to collect the clinical data.The normal control group was taken from the healthy subjects(no tumor,severe acute and chronic infection and other life-threatening diseases);Esophageal cancer patients from the First Affiliated Hospital of Guangxi Medical University thoracic esophageal cancer patients(esophageal cancer cases diagnosed with endoscopic pathology biopsy or postoperative pathological examination results).In the second step,the miRNAs in esophageal cancer patients and normal control group were extracted by TRIzol method.The levels of serum miRNA-210 were detected by real-time fluorescence quantitative PCR.In the third part,the relationship between the miR-210 level and the miRNA-210 in the observation group was compared with the clinical features.RESULT Observation group the miRNA-210,higher than the control group(P < 0.05),has nothing to do with the gender,age,pathological type,sex(P >0.05),is associated with TNM stage,histological grade,lymph node metastasis(P < 0.05),stage ? above ?,stage ? above(?-?)(P < 0.05),differentiation of high above middle,differentiation is higher than in low differentiation(P < 0.05).CONCLUSIONS(1)The expression of miR-210 in esophageal cancer patients was higher than that in normal subjects,suggesting that the elevation of miR-210 was related to the development of esophageal carcinoma.May inhibit tumor suppressor gene,leading to the occurrence of esophageal cancer.(2)The serum miR-210 in patients with esophageal cancer was associated with TNM staging,histological grade and lymph node metastasis,which was not related to pathological type.The higher the malignancy of esophageal cancer patients,the higher the level of miRNA-210 expression.
