Objective: To investigate the expression levels of MMP-2,E-cadherin and Suvivin in ESCC and to explore the correlations between these factors and ESCC development.Methods: A total of 156 patients with ESCC were enrolled in this study,including 121 cases of initial onset,35 cases of recurrence of patients.Tumor tissue and esophageal normal mucosal tissue were collected and esophageal normal mucosal tissues were at least 5 cm away from tumor tissues.The expression of MMP-2,E-cadherin and Suvivin in ESCC were measured by immunohistochemistry(S-P method).Results: 1.MMP-2 was mainly expressed in the extracellular and cytoplasm of normal esophageal squamous cells,while it was mainly concentrated in the cell membrane of ESCC cells.The expression of E-cadherin in esophageal squamous cell carcinoma cells did not change,and the expression position of E-cadherin was concentrated in the cell membrane,but the expression level was decreased.The expression level of Suvivin in normal esophageal squamous cells was low,but it was well expressed in ESCC cells.And the expression position was mainly concentrated in the nucleus and cytoplasm,but mainly in the cytoplasm.2.The positive expression rate of MMP-2 in poorly differentiated degree(73.68%)and moderate differentiation degree(66.33%)tumor tissues was significantly higher than that in the highly differentiated tumor tissues(38.89%)(P < 0.05).And its positive expression rate in deep infiltration depth tumor tissues(74.22%)was significantly higher than that in shallow tumor tissues(38.98%)(P < 0.05).The positive expression rate of MMP-2 in tumor tissues of patients with lymph node metastasis(85.36%)was significantly higher than that in tumor tissue of patients without lymph node metastasis(52.17%)(P < 0.05).The expression rate of E-cadherin in poorly differentiated tumor tissues(26.31%)was significantly lower than that in the highly differentiated degree(55.56%)and moderate differentiation degree(45.54%)tumor tissues(P < 0.05).And its positive expression rate in deep infiltration depth tumor tissues(30.93%)was significantly lower than that in shallow tumor tissues(61.02%)(P < 0.05).The positive expression rate of E-cadherin in tumor tissues of patients with lymph node metastasis(24.39%)was significantly lower than that in tumor tissue of patients without lymph node metastasis(53.04%)(P < 0.05).The expression rate of Suvivin in poorly differentiated tumor tissues(84.21%)was significantly higher than that in the highly differentiated degree(38.89%)and moderate differentiation degree(69.31%)tumor tissues(P < 0.05).And its positive expression rate in deep infiltration depth tumor tissues(75.25%)was significantly higher than that in shallow tumor tissues(45.76%)(P < 0.05).The positive expression rate of Suvivin in tumor tissues of patients with lymph node metastasis(87.80%)was significantly higher than that in tumor tissue of patients without lymph node metastasis(56.52%)(P < 0.05).In addition,tumor size,gender and age had no significant difference in the expression of MMP-2,E-cadherin and Suvivin(P > 0.05).3.The positive expression rate of MMP-2 in tumor tissue of recurrent patients(85.71%)was significantly higher than that in tumor tissues of initial onset patients(59.50%)(P <0.05).The positive expression rate of E-cadherin in tumor tissue of initial onset patients(73.55%)was significantly higher than that in in tumor tissues of recurrent patients(31.42%)(P < 0.05).The positive expression rate of Suvivin in tumor tissue of recurrent patients(82.85%)was significantly higher than that in tumor tissues of initial onset patients(58.67%)(P < 0.05).4.In ESCC tumor tissues,there was a positive correlation between the expression of MMP-2 and the expression of Suvivin,while there was a negative correlation between the expression of E-cadherin and the expression of Suvivin.Conclusions:The expression position and expression level of MMP-2,E-cadherin and Suvivin in ESCC tumor tissues were significantly different from that in normal esophageal mucosa.They can be used as an effective means of diagnosis of ESCC.
