Objectives Using Meta analysis to explore the association between HLA-? gene polymorphisms and helicobacter pylori infection,proving that if any contact between them,revealing the genetic factors whether HLA-?genes has effect to H.pylori infection.Methods The data retrieval about Chinese and English database,including CNKI,Wan Fang,VIP,PUBMED,Chinese biomedical literature databases,MEDLINE,OVID,COCHRANE and other medical database.Time range from January 1995 to June 2016.Literature on the association between HLA-class II gene polymorphism and H.pylori infection.The retrieved literature was screened according to the inclusion and exclusion criteria.read the literature carefully after screening,then use data extraction and quality evaluation.the data extraction can be divided into H.pylori infection group(observation group)and H.pylori uninfected group(control group)which is need Meta analysis;Quality evaluation report is according to the STREGA specification,<3 was low quality literature.The effects of measurement results are I2,Z,P,OR and 95%CI.According to the forest map of each point,make the summary table of Meta analysis results.If necessary,use the sensitivity analysis,subgroup analysis.Funnel plot was used to evaluate the publication bias,and the Egger s method was used for quantitative evaluation.Results 1 A total of 16 gene loci were included in DQA1,Among the DQA1*0102 sites,the frequency of H.pylori infection group was lower than H.pylori uninfected group(P=0.0008,OR95%CI: 0.72(0.59,0.87)),that is,DQA1*0102 is a protective gene against H.pylori infection.The subgroup analysis by region showed that DQA1*0102 is a protective gene for H.pylori infection in Asia(P=0.03,OR95%CI: 0.68(0.49,0.96)),while in Europe it is no longer a protective gene(P=0.43).the frequency of H.pylori infection group was higher than H.pylori uninfected group(P=0.009,OR95%CI: 1.35(1.08,1.69)),that means,DQA1*0103 is a susceptibility gene for H.pylori infection.DQA1*0104 and0301 gene loci are susceptible genes of H.pylori infection(P=0.0002,OR95%CI: 1.95(1.37,2.76)),(P=0.04,OR95%CI: 1.32(1.01,1.71)).However,DQA1*0104(P=0.06)and 0301(P=0.11)loci are no longer susceptible genes of H.pylori infection after sensitivity analysis.2 A total of 19 gene loci were included in DQB1,Among the DQB1*03032 is a protective gene against H.pylori infection(P=0.0003,OR95%CI: 0.55(040,0.76)).And the literature was from Asia,so it is a protective gene for Asian populations.DQB1*04 and 0401 are susceptible genes of H.pylori infection(P=0.02,OR95%CI: 2.56(1.14,5.75)),(P=0.0004,OR95%CI: 1.87(1.32,2.65)).DQB1*0602 and 0604 sites are protective genes of H.pylori infection(P=0.02,OR95%CI: 0.69(0.50,0.94)),(P=0.0009,OR95%CI: 0.50(0.30,0.84)).However,DQB1*0602,0604 was no longer statistically significant(P=0.08),(P=0.24).3A total of 38 gene loci were included in DRB1,Among the DRB1*08,11 Comparison between two groups,(P=0.03,OR95%CI: 1.61(1.05,2.49)),(P=0.04,OR95%CI:1.43(1.02,2.00)).DRB1*08 is a susceptibility gene for H.pylori infection.However,DRB1*08,11 is no longer the susceptibility gene of H.pylori infection(P=0.07),(P=0.10)after the sensitivity analysis.In addition,subgroup analysis was performed on the DRB1*11 locus(grouped by H.pylori diagnostic method).Whether the DRB1*11 site is associated with H.pylori infection,which used a number of trials to determine,more data will be needed(P=0.05,OR95%CI: 2.33(1.01,5.39)).Among the DRB1*12,there is no statistical difference between the two groups in DRB1*12(P=0.28).However,subgroup analysis(by Region)showed that DRB1*12 was a protective gene for H.pylori infection in Asia(P=0.04,OR95%CI: 0.45(0.21,0.96)).Whether DRB1*12 is associated with H.pylori infection in Europe,more data will be needed(P=0.05,OR95%CI: 2.62(0.99,6.90)).Subgroup analysis of DRB1*1502 loci(by Region),Whether DRB1*1502 is associated with H.pylori infection in Europe,more data will be needed(P=0.05,OR95%CI: 3.15(0.98,10.15)).Conclusions 1 DQA1*0102 is associated with H.pylori infection,which is a protective gene against H.pylori infection in Asian populations.2 DQA1*0103 is associated with H.pylori infection,which is a susceptibility gene for H.pylori infection.3 DQB1*04 is associated with H.pylori infection,which is a susceptibility gene for H.pylori infection.4 is associated with H.pylori infection,which DQB1*0401 is a susceptibility gene for H.pylori infection.5 DQB1*03032 is a protective gene against H.pylori infection,and is a protective gene for Asian populations.6 There is no clear correlation between DRB1 and H.pylori infection.
