AIM:To investigate the prevalence of Helicobacter pylori(Hp) infection and the strains of Helicobacter pylori in the family of children with recurred abdominal pain.To explore human leukocyte antigen DQB1 and DRB1 allele frequencies in these family.To investigate the relationship between clinical outcome and human leukocyte antigen DQB1 and DRB1 alleles after Hp infection, so that we can find if there are some associated genes affected Hp infection.METHODS:The serum Hp antibody (Ig-G) is tested by dot immunogold filtration assay and the Hp immunophenotyping is determined by Western blot (immunoblot) technique among 118 members in 20 families of children with recurred abdominal pain.There are 118 members with Hp infection in this family. PCR-SSP technique is applied to identify HLA-DQB1 and DRB1 allelic distribution between 69 members with recurred abdominal pain and 49 members without recurred abdominal pain.RESULT: The total seropositive rate in 118 members are 100% and the Hp immunophenotyping antibodies are 96.6%.The prevalence of Hp I typeis 55.1%.The prevalence of Hp typeⅡis 41.5%. There is significant difference between children and elders group about Hp strains rate(P<0.05).HLA- DQB1*09, DRB1*09 allelic frequency is higher in the members with recurred abdominal pain than that in the members without recurred abdominal pain, but the signification all disappeared when the allelic Bonferroni correction for comparisons were applied as required(14%vs6%, X2= 4.012, P=0.045, Pc<0.05; 10%vs5%, x2=5.636, P=0.018, Pc>0.05, respectively)。CONCLUSION:The seropositive rate of Hp antibodiies is high in the family with recurred abdominal pain. There is familial cluster phenomenon in family members suffering from recurred abdominal pain.lt should be studied further whether HLA-DQB1*09 and DRB1*09 allele are associated with abdominal pain.
